飲食成份對細胞色素P450的影響

Modulation of Cytochrome P450s by Dietary Components

摘要


隨著社會工業化,日常生活中接觸與攝取到一些毒性物質的機會增加,這些脂溶性外來物在動物體內可經由解毒酵素系統中的各種酵素反應,包括氧化、還原、水解、氫化以及去烷基等反應,將其轉變成較易排出的水溶性狀態。細胞色素P450(cytonchrome P450, CYP)為phase l解毒系統反應中負責外來物,例如藥物、致癌物、食品添加物及環境污染物等之氧化反應的酵素,在解毒酵素系統中佔有重要的角色。一些細胞內的CYP酵素,可受到一些化學物質,如多環狀芳香族碳氫化合物,巴比妥,乙醇類等物質的誘發。而其含量與表現亦會受到生理、營養素或飲食因子的調控,例如肥胖、禁食、飢餓、醣類、蛋白質、脂質、維生素、礦物質、有機硫化合物、多酚類化合物或食品添加物等。營養狀況會影響動物體內代謝毒物的能力,而毒物也會影響體內的營養素狀況,良好的營養狀況才能有效的將有毒的外來物泄出去,在本文中將主要針對飲食成份對CYP的調控作詳細的討論。

Xenobiotic metabolizing enzymes are involved in the biotransformation of lipophilic xenobiotics, including carcinogens, into more polar metabolites for excretion. Cytochrome P450 (CYP)-dependent monooxygenases are a family of phase l xenobiotic metabolizing enzymes that introduce a functional group such as -OH into the substrate. Some CYPs are inducible by various chemicals, such as polycyclic aromatic hydrocarbons, Phenobarbital and ethanol etc.. In addition, the expression of CYPs can also be modulated by various physiological conditions, nutrients and dietary components, including obesity, fasting, carbohydrate, protein, fat, vitamins, minerals, organosulfur compounds, polyphenols and food additives. Therefore, the nutritional status and the xenobiotic metabolizing system are closely associated. In this review, their relationship is discussed.
