DNA Fingerprintings of Five Chinese Herbal Medicines Revealed by RAPD Analysis

五種中藥材去氧核醣核酸指紋分析圖譜的建立

摘要


本試驗選取五種根莖類生藥材:柴胡、龍膽、白朮、黄連及當歸,進行逢機擴增多型性DNA(random amplified polymorphic DNA, RAPD)的反應,以期建立中藥材去氧核醣核酸指紋分析圖譜,作爲市售中藥材基源鑑定之用。本研究中以一條包含十個任意核苷酸序列的引子(primer)0PP-07:5'GTCCATGCCA3',對上述五種藥材所抽取之DNA進行RAPD分析。試驗結果分別得到各生藥之特定電泳圖譜。今後將針對其不同來源的真僞品,進行比對分析,並篩選反應效果良好的引子,以達快速鑑定的目的,其結果預期將可作爲中藥品質管制的參考。

The RAPD (random amplified polymorphic DNA) technique was employed to construct the fingerprints of five Chinese herbal medicines, Radix Bupleuri Chinensis, Radix Gentianae, Rhizoma Atractylodis Macrocephalae, Phizoma Coptidis, and Radix Angelicae Sinensis, all collected from stores in Taipei. In the present study, a decamer primer OPP-07:5'GTCCATGCCA3' was used in RAPD analysis to reveal the special fingerprints of the live herbal medicines mentioned above. The aim of the study is to provide a fast, easy tool to differentiate the Chinese herbs from their adulterants. The technique will contribute to the quality control of imported Chinese herbs.
