台灣地區環頸雉遺傳組成多樣性和族群遺傳結構

Genetic Diversity and Population Genetic Structure of the Ring-necked Pheasant (Phasianus colchicus) in Taiwan

摘要


採集自野外和人工養殖的環頸雉(Phasianus colchicus)共97隻個體,分析長度464bp的粒線體控制區DNA序列,發現有24個變異位置,皆為鹼基替代(substitution),可以定義出13個不同的基因型(haplotypes)。新建構之演化親緣關係樹主要分成兩群:基因型p13和已發表的高麗雉基因型為一群,另外基因型p10~p09為一群。從基因型在台灣各環頸雉族群間的分布情形,可以區分出外來環頸雉和台灣環頸雉的特有粒線體DNA基因型。估算台灣本島野外四個環頸雉族群的族群內遺傳多樣性指數、族群間的遺傳分化指數和基因交流程度發現:台東池上及台中清泉崗族群的族群內遺傳多樣性指數最低,分別為(h = 0.257±0.142;π = 0.00431±0.00294)及(h = 0.325±0.125;π = 0.00140±0.00054)。族群間遺傳分化指數最低與基因交流程度最高的族群是台南和花蓮鳳林族群(Nst= 0.17802;Nm=2.31)。

Samples of 80 and 17 ring-necked pheasants (Phasianus colchicus) were collected from the wild and domestic farms, respectively for an investigation of their genetic make-up. A fragment of 464bp of the control region in the mitochondrial genome was chosen for the analysis. Twenty-four variable sites, which are all nucleotide substitutions, can be defined by 13 haplotypes. The reconstructed relationship of haplotype phylogenies can be categorized into two discernible groups, namely, halotype P13 (same as the P.c. karpowi) and haplotypes P01 to P09. The haplotypes can be used to distinguish the Taiwan ring-necked pheasant and imported ring-necked pheasant. The groups of Taitung Vhihshang (h=0.257±0.142, π=0.00431±0.00294)and Taichung CCK airport (h=0.325±0.125, π=0.00140±0.00054)had the lowest genetic diversities. The genetic divergence was lowest and the gene flow was highest between groups of Tainan and Hualien Fenglin (Nst=0.17802;Nm=2.31).
