臺灣產枇杷屬之遺傳變異分析

An assessment of genetic variation of the genus Eriobotrya in Taiwan

本研究採用逢機擴大多型性去氧核醣核酸方法,研究臺灣原生之枇杷屬植物族群之遺傳變異,共分成四個區域,10個族群共144個樣本進行研究。共使用13個引子產生186個條帶其中單型性者有106個,單型性占56.99%,多型性條帶有80個,多型性條帶佔43.01%,山枇杷屬各族群開花物候期有重疊,並無明顯生殖隔離的情況發生。主座標分析(PCA)及歸群分析後顯示鵝鑾鼻、赤牛嶺及墾丁公園屬於恆春山枇杷型;陽明山群,大禹嶺群,梨山群及天祥群分為臺灣山枇杷型;武威山枇杷型因遺傳距離幾乎與枇杷難以區分,分類處理應歸類為變種。AMOVA分析結果,區域間變方成分百分比是11.18%,地理區域內族群間的變方成分百分比是41.47%,族群內個體間的變方成分百分比是47.35%。恆春山枇杷型之平均基因歧異度及多型性條帶比例高於其它區域,顯示其較高分化程度,平均遺傳歧異度達0.338,基因流(Nm)數值為0.9791。

Genetic differences among native populations of Eriobotrya deflaxa in Taiwan were investigated in order to shed light on the microevolution and intraspecific taxonomic relationships of the species. The commonly cultivated E. japonica was used as an outgroup. In this study, 144 individual samples from nine populations in four geographic areas were evaluated using RAPD markers. Thirteen RAPD primers were used and 186 RAPD markers were scored. Among these markers, 106 (56.99%) were monomorphic and 80 (43.01 %) were polymorphic. The results of the UPGMA cluster analysis based on polymorphic RAPDs revealed that three major groups could be recognized, i.e., Yang-Ming Shan, Da-Yu-Ling, and Tian-Siang belonged to E. deflexa form. deflexa; Eluanbi, Chi-Niu Ling, and Kenting Park belong to E. deflexa form. koshunensis; and N. Dawu Shan, and Dahan Shan belonged to Eriobotrya deflexa var. buisanensis. E. deflexa var. buisanensis was the most distinct on the clustering tree and was linked to E. japonica. However, the result of a principal coordinate analysis showed that E. deflexa var. buisanensis was closer to other members of E. deflexa than to E. japonica. Analysis of molecular variance (AMOVA) revealed that that the variance components among regions, among populations within regions, and among individual samples within populations were 11.18%,41.47%, and 47.35% of the total variance, respectively. The average G_(st) value was 0.338, and the Nm gene flow index was 0.979.
