應用花粉管基因導入法輔助甘藍抗黑腐病育種之研究

Studies on the Application of Pollen Tube Gene Transfer Method to Black Rot-Resistant Cabbage Breeding

摘要


為了探討應用花粉管基因導入法輔助甘藍抗病育種之可行性、本研究以抗黑腐病之甘藍品種P1281552為外源DNA供給親(donor plant)商業食用品種“初秋”為接受親(recipient plant),以花粉管基因導入法,進行甘藍之苦因轉殖。經檢定轉殖處理之T1植株,237株中存在56株(27/137株及29/100株)呈抗病性反應,並以RAPD分析,證實外源DNA可順利進入接受親。

Introduction of black rot resistance was carried out by transferring total genomic DNA from black rot resistance variety P1281552 into the edible 'KY cross' cabbage via pollen lube gene transfer method, Among 237 iudividuals evaluated by cut-leaf inoculation, 56 showed resistant to blank rot. Some of these resistant plants with the phenotype of 'KY cross' can be used as materials for further breeding. Furthermore, random amplified polymorphic DNA markers were found useful to distinguish the difference between parents and transgenic progeny.
