無機鹽類濃度及有機添加物對文心蘭擬原球體及組培苗生長之影響

Effect of Salt Strength and Organic Additives on the in vitro Growth of Protocorm-Like-Bodies and Plantlets of Oncidium Gower Ramsey

摘要


本試驗以文心蘭(Oncidium Gower Ramsey)兩品種之花梗節芽誘導PLBs及芽體,並建立微體繁殖體系。取直徑約0.5公分之PLBs,將其置於含0.4mg/L BA及0.1mg/L NAA之Murashige and Skoog (MS)及花寶一號培養基,及不同濃度的鹽類及有機添加物,以探討適合擬原球體(protocorm-like-body, PLBs)增殖之培養基。Onc. Gower Ramsey 'Hamana'之PLBs培養於以MS鹽類爲主的培養基,其增殖率在全量及半量的MS培養基較高;以花寶主的培養基中,含2g/L較1g/L培養基的效果較佳。在不同有機添加物對Onc. Gower Ramsey PLBs的生長試驗上,以含有3g/L tryptone之半量MS培養基的效果較佳。另外取高約1-2公分,帶3-4片葉片的分化小苗,培養於含不同濃度有機添加物(香蕉、胡蘿蔔、馬鈴薯、椰子水、及tryptone)的半量MS培養基,以探討最適合文心蘭瓶苗生長的培養基,試驗結果顯示,在同時含香蕉、馬鈴薯及tryptone三種成份的培養基生長的小植株不論在株高、葉數、根數、根長、鮮重、及乾重均較其他培養基爲佳;不含添加物之1/2MS培養基、及含香蕉的培養基次之,而在含馬鈴薯之培養基較差。鮮重的增加倍數以在1/2MS培養基的效果較佳。

The micropropagation system of the oncidium orchid was established, the flower stalk of two clones of Oncodium Gower Ramsey was used for inducing protocorm-like bodies and shoots. PLBs, including partial differentiated shoots, were cultured on Murashige and Skoog (MS) and organic additives were tested for optimal comditions of multiplication. PLBs of Onc. Gower Ramsey 'Hamana' were best multiplied on full strength and half strength MS media. For the Hyponex-based salts, PLBs grew more vigorous on 2g/L Hyponex medium with or without growth regulators. In the experiment of organic additives, the PLBs of Onc. Gower Ramsey grew better on Tryptone medium. The growth of plantlets with the height of 1-2 cm and 3-4 leaves was compared on MS basal medium containing various organic compounds, including banana, carrot, potato, coconut water, and tryptone. The result showed that the growth of oncidium plantlets was the best on medium containing banana, potato and tryptone altogether with respect to their plant height, leave number, root number, root length, fresh weight, and dry weight. The next better media were the control and the Banana medium, followed by the Potato medium. The fresth weight increment was higher on the control medium than others.
