台灣和尚蟹甲殼天然幾丁聚醣皮膜與人工合成幾丁聚醣薄膜之物性研究

Investigating the Physical Properties of Natural Chitosan Membrane from Carapaces of Taiwanese Soldier Crabs, Mictyris brevidactylus and Artificial Chitosan Membranes

摘要


台灣種短趾和尚蟹(Mictyris brevidactylus)由新竹香山濕地(GPS座標:N24°46' 54.9”; E120°54' 52.5”)捕捉,經酸(1 N HCl)、鹼(1 N NaOH)處理後,從和尚蟹甲殼部位取得天然幾丁聚醣皮膜。另由市售幾丁聚醣粉末進行薄膜再造程序(membranecasting),取得人工合成幾丁聚醣薄膜。本研究針對幾丁聚醣膜之去乙醯率(deacetylation degree)、皮膜厚度、分子量、含水率、拉力強度以及形變伸長量等物理性質進行試驗並作交互比對分析。 結果發現,和尚蟹幾丁聚醣皮膜在去乙醯率到達48%-55%範圍時,具有最大皮膜厚度與最小拉力強度。去乙醯率低於48%-55%區間時,皮膜厚度隨去乙醯率增加而變厚(但皮膜強度卻逐漸減弱);相反地,去乙醯率高於48%-55%區間時,皮膜厚度隨去乙醯率增加而變薄(但皮膜強度卻逐漸增強)。在不同去乙醯率條件下,皮膜厚度與強度之間呈現高負相關趨勢(R^2=0.9547),但皮膜含水率與皮膜厚度之間卻呈現高度正相關趨勢(R^2=0.8665)。 而人工合成之乾燥幾丁聚醣薄膜厚度與幾丁聚醣溶液濃度呈現正相關。但吸水澎潤後,除5mg/mL-8mg/mL濃度條件範圍下所合成之人工薄膜仍維持足夠的結構強度外,其餘條件下之人工幾丁聚醣薄膜皆於澎潤過程中吸水瓦解。

Relationships between deacetylation degree (DD), membrane thickness, moisture and tensile strength of the natural chitosan membranes purified from Taiwanese soldier crabs (Mictyris brevidactylus) were investigated. The artificial chitosan membranes casted from commercially available chitosan powders were also prepared and compared. The results revealed that the maximal membrane thickness and the minimal tensile strength of the natural chitosan membranes were found while DD value approached a range of 48%-55%. When DD value was lower than that range, membrane thickness increased and tensile strength decreased with the DD value. Membrane thickness decreased and tensile strength increased with increasing DD value as the range went higher. No matter how the DD value changed, a significant negative correlation between membrane thickness and tensile strength was observed (R^2=0.9547); the membrane thickness and moisture were well correlated (R^2=0.8665). On the other hand, the thickness of the artificial chitosan membrane was positively dependent on the concentration of chitosan solution during membrane casting procedure. After adequately swelling the membranes by deionized water, all of them were dissolved except those caste from the concentrations of chitosan solution from 5mg/mL to 8mg/mL.
