不同特性根瘤菌及菌根菌接種對雜交相思樹組織培養苗生長之效應

Effects of Various Rhizobium Sources and Coinoculation with Mycorrhizal Fungi on the Growth of Tissue-Cultureed Acacia mangium X auriculiformis Hybrid Plantlets

摘要


本研究係以微體繁殖生產之直幹與耳莢雜交相思樹培植體為宿主,接種根瘤菌及菌根菌,探討不同宿主及產酸、產鹼特性之根瘤菌及菌根菌接種之效果。試驗採三因子參試之逢機完全區集設計之複因子試驗,以根瘤菌原宿主為第一因子,次為菌株之產酸或產鹼特性,第三因子則為接種菌根菌與否。結果顯示,單獨接種根瘤菌及雙重接種菌根菌及根瘤菌,對苗木生長明顯優於僅接種菌根菌與完全沒有接種之對照處理。接種來自不同宿主之根瘤菌,經觀測苗木生長性狀及共生特性,以來自直幹相思樹之菌株最佳,接種植株產生之根瘤數約為來自耳莢及台灣相思樹之根瘤菌之3-4倍,獲悉根瘤菌共生效應,相思樹之雜交母本具有若干程度之遺傳控制。產鹼根瘤菌對苗木之結瘤數於酸性介質為產酸根瘤菌之2.4倍,培育介質偏酸性且所施缺氮營養液亦為酸性,導致產鹼根瘤菌較適應此種環境。菌根菌加根瘤菌雙重接種以及單獨接種根瘤菌,對苗木生長之效應以單獨接種根瘤菌最好。所施之缺氮營養液其含磷量充足以及先接種菌根菌再接種根瘤菌,兩個作業方式造成此結果。試驗結果也顯示根瘤數與葉部含氮量呈極顯著相關,即根瘤數量愈多,含氮量愈高。葉面積與葉部含氮量間,葉面積與苗木之高生長、葉乾重、莖乾重以及根瘤活性與葉面積間皆呈極顯著相關。

The objectives of this research were to verify if there is a difference in growth performances and nodule properties among tissue-cultured plantlets of Acacia mangium X auriculiformis hybrid that were inoculated with the 6 Rhizobium strains separately and a mixed inoculum of VA (vesjcular arbuscular) mycorrhizal fungi commonly. In a factorial inoculation experiment, 6 Rhizobium strains were categorized by their acidproducing or alkaline-producing and their host species. The growth of plantlets which were inoculated with Rhizobium alone and both with Rhizobium and VA mycorrhizal fungi were better than the inoculation with mycorrhizal alone and untreatment. Plantlets which were inoculated with Rhizobium strains isolated from A. mangium were dominant to others in growth performances and symbiotic characteristics. The number of nodules were 3 to 4 times than isolated from A. auriculiformis and A. confusa. This implics that Rhizobium strains are host-specific and symbiosis is controlled genetically by maternal compatibility of host plants. Alkaline-producing strains were 2.4 times than acid-producing strains for nodule numbers. Alkaline-producing strains were more effective to symbiosis with plantlets than acid-producing strains. One explanation is that growth medium of plantlets was weak acidity and non-nitrogen Hoagland solution fertilized during the period of experiment was acid, too, causing a poor growth condition for acid-producing strains. Single inoculated with Rhizobium was superior to dual inoculation of Rhizobium and mycorrhiza. However, single inoculation of Rhizobium gave significantly higher growth, nodulation over the rest of the treatments. This may result from time-lag inoculation and caused by use of non-nitrogen ferlilizer. In correlation analyses of growth characteristics, nodule number and leaf total nitrogen were significantly correlated. The more nodule formed, the more nitrogen fixed. There is a significant correlation between leaf area and leaf nitrogen content. Leaf area was significantly correlated with plantlet height, leaf dry weight and stern dry weight. The correlation of nitrogenase activity (acetylene reduction) and leaf area was significant too.
