Enhanced Expression of C-Ki-Ras Oncogene and High Serum Level of Total Lactic Dehydrogenase in a Papillary Adenocarcinoma of the Ovary

A 57-year old woman was found with a stage III ovarian cancer. Cytoreductive .surgery was done. A preoperative blood sample was obtained for determination of lactic dehydrogenase (LDH). An ovarian tissue specimen was taken for determination of expression of c-Ki-ras oncogene. Serum total LDH level was measured by a blood chemistry automatic analyzer. LDH isozymes activities were determined by electrophretic separation on cellulose acetate membrane. The total LDH level was 2562 IU/ml. The isozymes of LDH-1 to LDH-5 were 21.5%, 35.1%, 20.4%, 9.4% and 13.6, respectively. A 15-fold increase ofc-Ki-ras RNA transcripts was demonstrated. The concomitant elevation of total LDH level and expression of c-Ki-ras may suggest that enhanced expression of ras oncogene results in increased synthesis of p21, which may then transactivate the LDH genes at the transcriptional level.
