Verification and Analysis of the Result of the Yeast Two Hybrid Screens for Rotini, a Drosophila Golgi Protein

新近發現的果蠅高基氏體蛋白rotini可經由細胞膜上的多醣蛋白HSPGs調控果蠅的形態基因。rotini基因是第一個被搜尋辨識出來,對於特殊母源效應是致死基因,且已有人支持它是體節極性基因(segment polarity gene)中的一個新成員。在rotini突變的果蠅品系中,其Hedgehog及HSPGs之抗體染色結果極具戲劇化地下降。rotini和人類的高基氏體蛋白GPP34系列同源,後者被報導和細胞內運輸有關。爲了暸解rotini的功能,我們進一步進行雙雜交搜尋,並得到27個可能的候選基因;經過確認與分析之後,發現有一個具v-SNARE活性者可提供我們對於rotini的功能有更進一步的認識。

Rotini, a novel Golgi specific protein, regulates distribution of morphgens through HSPGs. Rotini is first identified in screens for lethal gene with specific maternal effect, and has proven that is a new member of segment polarity gene. In rotini mutant clone, the antibody staining of Hedgehog and HSPGs decrease dramatically. Rotini is the homolog of human Golgi GPP34 family, which is reported that is involved in the intracellular trafficking. In order to approach the function of rotini, we preceded the yeast two hybrid screens, and got 27 candidates. After verification and analysis, there is a gene with v-SNARE activity which provides us more hit for the function of rotini.
