番茄枝科植物對青枯病菌之反應

Reaction of Some Annonaceous Plants to the Bacterial Wilt Pathogen

摘要


番荔枝、牛心梨、刺番荔枝、山刺番荔枝圓滑番荔枝、奇美釋迦香水樹及瓣立櫻林果等八種番荔枝科苗木中,以番荔枝及牛心犁對青枯病菌最感病,而圓滑番荔枝最具抗性。圓滑番荔枝二年生或3.5個月株齡之苗木,以莖部接種時,經二個月後外觀均無病徵,僅接種部位有維管束褐化現象;3-5個月大的幼苗以土壤混菌接種後二個月,外觀亦健康,雖有些植株之根部有病菌存在,但莖部均無掃描式電子顯微鏡之觀察,感病的番荔枝在莖頂接種青枯病菌後48-96小時,在接種部位下3公分之莖部導管內可見到細菌充塞,接種後144小時,導管內細菌量增加且向外湧出至鄰近細胞,有些導管已被瓦解並釋出大量的細菌,而抗病的圓滑番荔枝,在接種後48-192小時期間進行觀察,除極少數導管偶而可見到細菌外,其餘均無細菌存在。

Eight annonaceous plants (Annona squamosa A. reticulata, A. muricata, A. montana, A. glabra, a hybrid of A. squamosa x A. cherimola, Cananga odorata and Rollinia deliciosa) were tested for resistance to Pseudomonas solanacearum, the bacterial wilt pathogen of sugar apple. A. squamosa (sugar apple) and A. reticulata (bullock's heart) were most susceptible, while A. glabra (pond apple) was most resistant to the pathogen. stems of pond apple plants (2-year-old or 3-5-month-old) inoculated with P. solamacearum showed only vascular browning around the inoculated site, and appeared to be healthy after two months. When the 3-5-month- old seedlings of pond apple were transplanted into the infested soil, they also remained symptomless, though P. solanacearum in low number was detected in roots of some plants, but was not detected in stems of all plants tested. Sugar apple and pond apple seedlings inoculated by clipping off the stem apex by the infested scissors were examined by scanning electron microscopy. On susceptible sugar apple, numerous bacteria occluding the xylem vessels could be observed in the 3-cm stem segment below the cut end 48 and 96 hrs after inoculation; the number of bacteria was increased, and some vessels were disrupted, releasing a mass of bacteria to the nearby 144 hrs after inoculation. On resistant pond apple, however, nearly all vessels at the sections 0.2-3.0 cm below the cut end were absence of bacteria, occasionally bacteria were observed in only a few vessels during the 192 hr observation period.
