細葉山茶樹種抽出物之抗氧化性質探討

採集細葉山茶(Camellia tenuifolia (Hay.) Cohen-Stuart)之葉、花及果皮等不同部位試材,使用乙醇溶劑(75%)萃取各試材,所得固體抽出物含量大約在15~25%左右。應用Folin-Ciocalteu試驗法測定各抽出物之酚類化合物總含量,結果顯示,葉、花及果皮等抽出物之總酚量在95~135mg GAE/g之範圍(GAE係gallic acid equivalent之簡稱,即“沒食子酸當量”)。其中葉材之總酚量略高於果皮,唯差異不大,而花材最少。比較不同試材抽出物溶液對於DPPH自由基之清除效應,得知葉和果皮溶液在50μg/ml濃度之清除效率均達到90%以上,優於相同濃度之花材溶液(60~65%)。測試各項試材抽出物對於MeLo之氧化抑制效應,顯示葉材略優於果皮,而再優於花材。再應用TBA檢測法探討各項抽出物對於亞油酸抗氧化性質之影響,果皮抽出物對於降低亞油酸氧化之醛類產物具有最顯著之效果,次為葉材,花材居末。上述試驗結果也證實抽出物之酚類成分總含量與抗氧化性質之間具有正相關性。

Samples of the leaves, flowers and peels from oil-tea tree (Camellia tenuifolia (Hay.) Cohen-Stuart) were extracted with a 75% ethanol solution. The extract contents were found to be about 15~25%. The total phenolic contents of the extracts from different samples were determined by the Folin-Ciocalteu method and were found to be in the range of 95~135mg gallic acid equivalent/g, with the respective content order as that of leaves ≧ peels > flowers. In addition, the DPPH free radical-scavenging rates of the extract solutions were observed to be greater than 90% for leaves and peels, superior to 60~65% of the flower extract at the same concentration (50μg/ml). The inhibition effects of extracts on MeLo oxidation revealed that the leaf extract presented the highest inhibition rate, slightly better than that of peel extract, while that of the flower extract was the lowest. Moreover, the test of linoleic acid oxidation detected with thiobarbituric acid revealed that the peel extract demonstrated the highest reduction of aldehyde production from linoleic oxidation, with leaf extract the next and flower extract the lowest. From these experimental results, a positive correlation between total phenolic contents and antioxidant properties of the extracts was also confirmed.
