素心蘭花莖培養之形態發生

In vitro Morphogenesis from Flower Stalk of Cymbidium ensifolium

摘要


素心蘭花莖培植體於含有不同濃度2,4-D配合TDZ之1/3 MS培養基中生長,顯示添加2,4-D(2, 4-Dichlorophenoxyacetic acid)配合TDZ處理能促進素心蘭花莖組織形態發生,且以上段花莖組織的形態發生率較下段花莖高。當中以含有0.5、2.0或4.0 mg/L2,4-D配合各濃度TDZ處理的上段花莖褐化率為0%,形態發生率為100%。並隨著TDZ處理濃度增加,芽體形成數明顯增加,然TDZ濃度高於0.5 mg/L時,芽體形成則減少。隨2,4-D和TDZ處理濃度增加,根莖形成數則明顯增加,然二者濃度分別超過2.0 mg/L和1.0 mg/L時,根莖形成數則減少。就花序分化而言,低濃度2,4-D處理中增加TDZ濃度,花序形成數增加,而隨著2,4-D處理濃度增加,花序形成數明顯減少。就瓶內開花數調查顯示,隨著2,4-D處理濃度增加,開花數增加,然當2,4-D處理濃度達4.0 mg/L時,開花數則明顯減少。而花莖培殖體則以添加0.5-1.0 mg/LTDZ配合2 mg/L2,4-D之1/3MS培養基能明顯誘導最多芽體形成。

Flower stalk explants of Cymbidium ensifolium were cultured on 1/3 MS medium containing different concentration of 2, 4-D and TDZ. Upper flower stalk explants cultured on 1/3 MS medium supplemented with different concentration 2, 4-D and TDZ showed higher morphogenesis than lower flower stalk. Maximum percentage of morphogenesis achieved at 1/3 MS medium supplemented with 2.0 mg/L 2, 4-D and l.0 mg/L TDZ, browning was decreased when the 2, 4-D concentration was increased. Maximum numbers of shoot formation was achieved at 1/3 MS medium supplemented with l.0 mg/L 2, 4-D and 0.5 mg/L TDZ. Increasing the concentration of TDZ above 0.5 mg/L, the shoot formation was decreased. The maximum percentage of rhizome were achieved at 1/3MS medium supplemented with 2, 4-D (l.0 mg/L) and TDZ (0.1 mg/L). The rhizome formation was decreased significantly when the 2, 4-D and TDZ concentration was increased. The maximum number of inflorescence was achieved at 1/3 MS medium supplemented with 0.5 mg/L TDZ. The maximum number of flower formation was achieved at 2.0 mg/L 2, 4-D and l.0 mg/L TDZ.
