利用ISSR分子標誌分析毛豆品種(系)間之遺傳歧異度

Genetic Diversity of Vegetable Soybean Varieties (lines) Based on ISSR DNA Markers

摘要


本研究是以高雄區農業改良場毛豆育種上常用之親本及重要育成品種(系)共80個為材料,利用簡單序列間重複(inter-simple sequence repeat, ISSR)標誌進行遺傳歧異度分析,以提升育種效率及培育更優良品種。由ISSR試驗結果顯示,100個引子中可篩選出具有多型性條帶之引子7個,分別為UBC-818、UBC-835、UBC-840、UBC-868、UBC-873、UBC-878、UBC-891,佔7%,以2個重複鹽基序列為主,其中又以具有ISSR3'-anchord的primer之擴增效果較佳。7個引子在80個毛豆品種(系)中共可擴增出60個DNA條帶,具多型性DNA條帶則有52條。其中以UBC868擴增出16個DNA條帶為最多,且於電泳膠片上900 bp處有一明顯的多型性條帶,此DNA條帶除了來自中國的Kuo China品系外,可視為常出現在黑色種皮毛豆中。利用UPGMA經集群分析結果,以遺傳距離係數0.41為分群依據,大致上可將黑色種皮毛豆與其他顏色種皮毛豆加以區分。

In order to improve breeding efficiency and accelerate the development of new vegetable soybean varieties, this study applied inter-simple sequence repeat (ISSR) to analyze the genetic diversity of vegetable soybean varieties (lines). Eighy samples consist of parents of breeding and breeding lines collected by the Kaohsiung District Agricultural Research and Extension Station were used for ISSR analysis. The results demonstrated that 7 out of 100 ISSR primers, including UBC-818, UBC-835, UBC-840, UBC-868, UBC-873, UBC-878, and UBC-891, showed polymorphic patterns. Most primers that amplified polymorphic bands belong to dinucleotide repeats with 3'-anchord sequences. Among 60 DNA fragments amplified by 7 primers, 52 bands showed polymorphism. The UBC-868 is the most efficient primer which generates 16 polymorphic bands, including the most significant 900 bp polymorphic DNA fragment. The 900 bp fragment is specifically observed in vegetable soybean varieties which have black seed coat, except the yellow seed coat variety Kuo China. Results from the UPGMA cluster analysis enable us to distinguish the soybean varieties with black seed coat from others, based on the genetic distance value of 0.41.
