含微量元素包裝水對LPS處理RAW264.7巨噬細胞抗發炎機制之探討

摘要


市售包裝水種類繁多,但是否具有抗氧化、抗發炎等保健效益,則極少有相關的研究。本研究是利用以LPS刺激RAW264.7細胞發炎模式,探討含微量元素的包裝水產品之抗發炎作用,分析(1)對細胞一氧化氮(NO)生成量。(2)細胞內誘發型一氧化氮合成酶(iNOS)蛋白質表現量。(3)細胞內COX-2蛋白質表現量。使用含微量元素包裝水200倍濃縮液原料,配製成相當成品濃度1,5,10,20倍組。試驗結果顯示:以MTT法檢測發現,含微量元素包裝水原料在五倍濃度劑量內,對細胞生長沒有影響;各濃度劑量均具有減少NO生成量及iNOS蛋白質表現量之作用,且呈現劑量反應。但COX-2表現量與控制組相當。由以上結果可知,含微量元素包裝水原料成份可能藉由抑制iNOS表現量,進而減少細胞NO生成。

There are many types of mineral water products in Taiwan. The anti-inflammatory effects of Secontaining mineral water were still unknown. The objective of the present study was to investigate the trace mineral containing water (including Se) products for their anti-inflammation effect on LPS-treated RAW 264.7 cells. The cell NO production, iNOS enzyme protein expression, and COX-2 enzyme protein expression were detected. The 200X stock of trace mineral containing water product was used and was diluted to 1X, 5X, 10X, and 20X. The cell growth curves were analyzed by MTT method, and it demonstrated that there were no influences by treating 1X and 5X stocks. However, it showed a dose-dependent decrease on NO product and iNOS protein expression by varied stocks treatments. The COX-2 protein expression had no differences to the control group. In conclusion, the stocks of trace mineral containing water products are able to inhibit the inflammation of RAW264.7 cells induced by LPS treatment, and it may be conducted the effect by decreasing iNOS protein expression and NO production.
