Detection of Genetic Variation in the VP2 Gene of Vaccine and Field Strains of Infectious Bursal Disease Viruses

野外株及疫苗株雞傳染性華氏囊病病毒第二病毒蛋白基因變異性分析

以反轉錄聚合酶鏈反應及限制酶分析法檢測25株台灣雞傳染性華民囊病(IBD) 病毒野外分離株與疫苗毒株第二蛋白基因之變異情形,結果發現以Taql、Sspl、Sacl、Nael、Hinfl、BstEII及Maelll等限制酶切割所得之圖譜在不同毒株間有所差異,並可基於此限制酶切割圖譜將25株毒株分成4個基因型,其中第1型包括3個亞型:即1A亞型(6株)包括第二血清型毒株、第一血清型傳統株、變異株及一株疫苗株,1B亞型及1C亞型分別包括2株及5株疫苗株;第二型則包含2 株疫苗株;第三型中3A亞型包括7株高病原性IBD(VVIBD)毒株台灣分離株,而3B亞型則包括1株日本VVIBD毒株;第四型則包括一株台灣VVIBD分離株及1株以色列VVIBD由來之減毒疫苗株。進一步以基因樹分析,結果顯示台灣VVIBD 毒株與法國及日本之VVIBD毒株極為相似。本研究結果顯示VVIBD毒株及其減毒疫苗株的VP2高度變異區,與傳統株、變異株、及傳統株之減毒痠苗株間有顯著之差異。

Reverse transcription - polymerase chain reaction of the VP2 gene followed by restriction endonuclease analysis was used to detect genetic variation among 25 strains of both field and vaccine strains of infectious bursal disease virus (IBDV) in Taiwan. Restriction fragment profiles generated by Taql, Sspl, Sacl, Nael, Hinfl, BstEII and Maelll showed variation between strains. The IBDV strains were placed into four molecular groups. Group 1 consisted of 3 subgroups. Subgroup 1A (6 strains) consisted of serotype II strains, classic serotype I strains, variant strains and one vaccine strain. Subgroups 1B and 1C consisted of 2 and 5 vaccine strains, respectively. Group 2 contained 2 vaccine strains. Group 3A contained 7 Taiwanese strains of very virulent IBDV (vvIBDV), while a Japanese vvlBDV strain was assigned to group 3B. Group 4 contained a Taiwanese vvlBDV strain and an Israeli vaccine strain (MB). A phylogenetic study indicated that Taiwanese vvlBDV strains are very similar to French and Japanese vvlBDV strains. It is concluded that hypervariable region of the VP2 of vvlBDVs and an attenuated derivative are different from classic strains, variant strains , and attenuated strains derived from classic strains.
