傳染性支氣管炎病毒株之抗原性分析

The Antigenicity Analysis of Infectious Bronchitis Viruses

摘要


為了探討不同雞傳染性支氣管炎病毒株之抗原性,本實驗針對分屬台灣Ⅰ型(TW-Ⅰ)與台灣Ⅱ型(TW-Ⅱ)之雞傳染性支氣管炎本土分離株以及大陸廣西省分離株,進行病毒增殖、蛋白質電泳與西方免疫轉漬試驗、交叉中和反應,以挑選出具有高病毒力價及廣泛抗原性的本土雞傳染性支氣管炎病毒毒株,供發展疫苗之用。在病毒力價方面,以A2296/95及A2575/98最高。在蛋白質電泳與西方免疫轉漬試驗方面,可以發現不同的血清型別特異性抗血清與各毒株間棘突結構蛋白之反應不同,其中與各病毒株間有較佳反應的血清型別為TW-Ⅰ之A1171/92、A1449/92及A2575/98。在交叉中和試驗方面,TW-Ⅰ中A1171/92及A1449/92血清與其他毒株中和力價百分比達71%。TW-II之間達64%-81%。因此TW-Ⅰ之A1171/92、A1449/92及A2575/98毒株可以供發展TW-Ⅰ疫苗,TW-Ⅱ的3個毒株在同一型別中皆具有相同的抗原性,A2296/95病毒力價最高,可以供發展TW-Ⅱ疫苗。

In order to select infectious bronchitis viruses (IBV) for vaccine development, several IBVs of Taiwan group Ⅰ (TW-Ⅰ) and Taiwan group Ⅱ (TW-Ⅱ), and one Guangxi isolate were tested by virus proliferation, Western blot, and cross neutralization tests. From virus propagation result, IBV strains A2296/95, and A2575/98 archived high virus titers after propagation. From Western blot result, the spike protein of A1171/92, A1449/92 and 2575/98 showed the highest reactivity. From the cross neutralization result, the cross reaction of A1171/92, A1449/92 reached 71% to other isolates and that of TW-Ⅱ isolates reached 64% to 81%. Therefore, A1171/92, A1449/92 and 2575/98 of TW-Ⅰ could be used for vaccine development. Since the three TW-Ⅱ isolates the same antigenicity, A2296/95 which archives the highest virus titer could be used for vaccine development
