以多引子聚合酶連鎖反應快速偵測台灣南部豬鏈球菌之血清型及毒力相關基因

Rapid Detection of Serotypes and Virulent factors in Southern Taiwan by Multiplex Polymerase Chain Reaction

摘要


豬鏈球菌(Streptococcue suis; S. suis)依據莢膜多醣體(capsular polysaccride; cps) 之不同已證實有35種不同血清型,其中第2型最常從發病豬中被分離出來。目前已知與本菌毒力相關的毒力因子有cps、溶血素(suilysin; sly)、細菌胞壁酸酶釋放蛋白(Muramidase-released protein; MRP)和細胞外因素(extracellular factor; EF)等。Suilysin具有細胞毒性能侵入到深層的組織,為高毒性毒力因子;而MRP 和EF都是分泌性蛋白,與本菌的毒性強度有關,MRP(上標 +)/EF(上標 +)為強毒株、MRP(上標 +) /EF(上標 *)為弱毒株、MRP(上標 -)/EF(上標 -)為無毒株。本研究綜合及毒力因子基因sly、mrp和ef之特異性引子對,以多引子聚合酶鏈鎖反應(multiplex PCR)快速鑑別分析本土分離株之血清型及毒力基因型,其中血清型第2 型佔54.5%,第1型佔27.3%基因型均為mrp(上標 +)/leƒ(上標 +)lsly(上標 +)。結果供建立本省南部豬鏈球菌分離株之基礎資料庫,並作為發展有效疫苗之重要參考依據,藉以防範此一人畜共同傳染病之入侵。

Streptococcus suis (S. suis) has been classified into 35 serotypes based on the structure of capsule polysaccharide (cps). Type 2 is the most frequently isolated serotype from infected piglets. Virulence factors possibly involved in the pathogenicity of S. suis, such as cps, suilysin (sly), muramidase-released protein (MRP) and extracellular factor (EF), have been identified. Suilysin, a highly virulent factor, has a cytotoxic effect and might facilitate a deeper penetration of S. suis into host tissues. Both MRP and EF are secreted protein and are associated with the high virulence of serotype 2 S. suis. Phenotype MRP(superscript +)/EF(superscript +), MRP(superscript +)/EF(superscript *), and MRP /FF- represent highly virulent, less virulent, and avirulent, respectively. In this study, we analyzed the genes of virulence factors (cps, sly, mrp and en by using multiplex PCR with specific primers to identify the strains of S. suis isolated from cases in the southern Taiwan. Results indicated that most strains belonged to serotype 2 (54.5%) and serotype 1 (27.3%). In addition, all serotype 1 isolates were mrp(superscript +)lef(superscript +)lsly(superscript +). The data are valuable to a more rapid diagnosis of S. suis and to the development of effective vaccines for controlling and eradicating this pathogen.
