The Complete Sequence of a Reticuloendotheliosis Virus Isolated from Chickens

由雞分離網狀內皮病病毒的全長基因

摘要


網狀內皮病病毒(REV)引起雞及數種禽類的生長不良、免疫抑制及慢性腫瘤的疾病。台灣的成年雞有95%的感染率。2005年7月於彰化-32週齡雄性白肉種雞血液分離出一株REV病毒,此原病毒全長序列為8,238 b.p.顯示典型反轉錄病毒的結構,前後各包括1段長末端重複序列,主要基因有gag,pol及env等,與插入禽痘病毒的REV病毒相似度高(98.7%),於env基因之穿膜部分含有免疫抑制基因,與其他REV比較以長末端重複序列變異最大,此序列可供為流行病學調查之用。

Reticuloendotheliosis virus (REV) causes runting, immunosuppression, and chronic neoplasia in a variety of birds, including chickens. Its serological prevalence reaches 95% in adult chickens in Taiwan. A REV from blood samples in a male 32-week-old broiler breeder farm in Changhua was isolated in July, 2005. The complete proviral sequence of this isolate was determined. Its proviral genome is 8,238 nucleotides in length and exhibits a genetic organization characteristic of replication-competent Gammaretroviruses. The provirus contains two identical long terminal repeats (LTR) and a complete set of genes including gag, poi and env. Alignments with other REV sequences showed high similarity. The whole gene of the present REV gene showed high nucleotide sequence identity with REV sequences inserted in fowl poxvirus (98.7%). Sequences coding for a previously reported immunosuppressive peptide contained in the transmembrane region of the env gene were well conserved in this REV. Comparing with reference REV strains, the LTR was the most divergent region, which could be use for epidemiological investigation.
