評估蛋雞餵飼幾丁聚醣之免疫調節

Evaluation the Immune Regulation of Laying Hens Fed by Chitosan

摘要


近年來由於抗菌劑的濫用引起許多的抗藥性問題,嚴重地影響到動物及人類的疾病控制,因此尋找其他的替代品成為目前相當重要的課題。本實驗主要研究幾丁聚醣配方以口服方式投與雞隻,了解其對於雞隻免疫反應之調控。以100ppm的濃度餵飼25週齡的蛋雞,分別測量餵飼前組(D0)、連續餵飼3天組(D3)、連續餵飼6天組(D6)以及D6組停止餵飼14天後觀察組(D20)血液中的IFN-γ及IL-2濃度變化;另外雞隻停飼14-20天後(S組)取其脾臟及腸道淋巴組織,觀察免疫激素的基因表現量變化。實驗結果發現D3組血中的IFN-γ濃度達到最高峰(3200.2pg/mL),並於D20組血中濃度降至180.2pg/mL;而IL-2的血中濃度則仍呈現持續上升的現象。此外,S組於脾臟中的免疫激素基因包括IL-1β、IL-6、IL-10、IFN-γ和K203,以及盲腸淋巴組織中的IL-6及IFN-γ之基因表現量相較未餵飼雞隻皆呈現下降之趨勢。綜觀各項結果,幾丁聚醣以口服餵飼雞隻,能夠引起周邊血液趨向T(下標 H)1路徑,有效地提高雞隻的免疫反應。然而淋巴組織則似乎有趨向減緩T(下標 H)1反應的傾向,惟需進一步的實驗證明。

The current overuse of antimicrobial agents is quite prevalent and may cause deleterious ramifications such as drug resistance or interruptions in human disease therapy. As levels of antimicrobial resistance continue to rise, it becomes ever more critical to locate alternative treatments. Chitosan, a deacetylated derivative from chitin, possesses antimicrobial properties partially due to its immune-regulatory functions. In the present study, the effect of chitosan on the immune response of chickens was investigated. Twenty-five week old laying hens were divided into three groups of where Group 1) was fed no chitosan (D0), Group 2) continuously fed 100 ppm of chitosan for 3 days (D3), and Group 3) continuously fed 100 ppm of chitosan for 6 days (D6). Hens belonging to the D6 group were euthanized at the 20th day (D20) after chitosan was discontinued on the 6th day. Serum (only for D20) and lymphoid tissue samples (S-group) were collected from the group D20 as well as from to the D3 group discontinued chitosan for 20 days to observe the cytokine and related gene expression profiles. An increase of serum IFN-γ was found in group D3 (3200.2 pg/mL) while it descended to 180.2 pg/mL in group D20. Serum IL-2 continued to elevate during the days when chitosan was fed. On the other hand, the gene expressions of IL-1β, IL-6, IL-10, IFN-γ, and K203 in the spleen were down-regulated and well as those of IL-6 and IFN-γ in the cecum lymphoid tissue. These results suggest that chitosan possesses immune regulatory effects due to the induced T(subscript H)1 responses in peripheral blood and the reduced responses in the lymphoid organs, however, further investigations are required to elucidate the distinct mechanism in detail.
