用免疫組織化學染色法分析龍葵水萃取物 抑制動脈粥狀硬化中脂肪斑塊蛋白之分析

摘要


動脈粥狀硬化(Atherosclerosis)是一個形成原因極為複雜的血管疾病,為冠狀動脈疾病及中風的重要致病機轉。平滑肌細胞由血管中層往內皮下間隙移行與增生是形成動脈硬化斑的重要步驟。根據東西方的醫學文獻報導指出,龍葵 (學名Solanum nigrum )具有抗氧化、抗發炎、降血脂與保護肝臟等醫學功能。本實驗以高脂肪飲食(high fat diet; HFD)餵食紐西蘭大白兔(New Zealand rabbit),並於飼料中添加0.25%、0.5%、1% 龍葵水萃取物 (Solanum nigrum water extract, SNWE) 。經十週後,取主動脈(aorta)一段,作切片 (section)與染色(H&E stain),並引用免疫組織化學染色 (immunohistochemical stain)經由電腦軟體定量及統計分析,偵測與細胞增生 (proliferation)有關的蛋白如: Ki-67,VEGF,HIF-1α; 細胞遷移 (migration)有關的蛋白如RhoA,Rac,cdc42 和 SDF-1α,CXCR4;及細胞凋亡 (apoptosis) 有關的蛋白Bcl-2,Bax表現的情形。結果發現SNWE可降低HFD誘引紐西蘭大白兔粥狀動脈硬化(Atherosclerosis)降低主動脈血管脂肪斑塊增生蛋白(Ki-67, VEGF, HIF-1α)和細胞遷移蛋白(RhoA ,Rac,Cdc42,和 CXCR4)的表現,降低抗凋亡蛋白(Bcl-2)、促進凋亡蛋白(Bax)的表現。 綜合H&E stain、免疫組織化學染色的結果,認為SNWE可以抑制動脈粥狀硬化中脂肪斑塊蛋白增生。降低HFD引起之心臟血管受損的程度而達到保護心血管的作用。對於未來保健食品的開發提供了更明確的目標。

Atherosclerosis, a complex and multifactorial disease, is the underlying mechanism for vascular disorders including coronary artery disease and stroke. Vascular smooth muscle cells (VSMC) proliferation and migration from media to subendothelial intima plays a major role in the lesion formation of atherosclerosis. The solanum nigrum water extracts (SNWE) have many medical functions including the antioxidant, antiinflamation, lowering lipid and protecting the liver, etc. In this study, the New Zealand Rabbits were given high fat diet with added 0.25%, 0.5% and 1% of SNWE. After ten weeks, a piece of the aorta of the rabbit was taken for a section, and H&E Stain as well as an Immunohistochemical Stain to detect the cell proliferation of proteins such as RhoA, Rac, cdc42, Bcl-2, Bax, HIF-1α, VEGF, SDF, CXCR4 and Ki-67. Results from H&E Stain and Immunohistochemical Stain assume that adding SNWE into diet is able to reduce the proliferation Proteins of the fatty streak in Atherosclerostic lesions. In conclusion, SNWE can protect cardiac vascular from damage caused by HFD. These results also provide a scientific prove for traditional herb on healthy-caring food.
