荷葉多酚萃取物抑制血管平滑肌細胞遷移之作用

摘要


動脈粥狀硬化是多數心血管疾病形成的必然過程,也是全球邁入老年化國家急欲解決的健康問題,其中以天然物來作為預防動脈粥狀硬化的研究方向越來越受到重視,以最天然無害又容易取得的天然物讓大眾在平時就能透過攝取來達到預防的效果,可以說是最有經濟效益的健康對策。目前已知血管平滑肌細胞移行至血管內膜是形成動脈粥狀硬化的重要過程,本實驗以台灣大量培育的大憨蓮荷葉為原料並萃取其多酚產物來處理A7r5細胞株觀察荷葉多酚萃取物對於血管平滑肌細胞遷移能力的影響。透過MTT assay、Wound healing assay、Boyden chamber assay、Gelatin Zymography assay、Western blot assay等方法來評估。在目前的實驗結果已知在不誘發細胞走向凋亡的劑量下,wound healing assay及Boyden chamber assay 的實驗中可知荷葉多酚能抑制細胞遷移,而Gelatin Zymography assay結果得知荷葉多酚能抑制MMP-2的活性。在western blot assay中NPEs可以降低血管平滑肌細胞的phospho-Akt的表現量以及ERK、p38的磷酸化作用來抑制細胞的遷移。

The atherosclerosis is the inevitable course during cardiovascular diseases progression. The health problem is the developed country urgent to solve, among them is being paid attention to the natural thing’s research direction of preventing the atherosclerosis. The natural thing easy to get that against atherosclerosis is the healthiest and most economic policy. Smooth muscle cell migrate to intimal is major processes causes the atherosclerosis. We extract polyphenol of Lotus leaf of Taiwan to prove that inhibitory effect on vascular smooth muscle cells (A7r5 cell line) migration. The MTT assay, Wound healing assay, Boyden chamber assay, Gelatin Zymography assay, Western blot assay…etc. are used to confirm. Present experimental results show NPEs can’t induce cell apoptosis but suppress hyperplasia of A7r5 cell in wound-healing assay; can suppress migration in Boyden chamber assay; can inhibit MMP-2 activity in Gelatin Zymography assay. In western blot assay shows NPEs can reduce the phospho-Akt expression in smooth muscle cell that also reduce phospho-ERK and phospho-p38 expression to suppress vascular smooth muscle cells migration.
