發光二極體光刺激對糖尿病老鼠燒燙 傷口癒合之影響

Effect of 630nm Light Emitting Diode Irradiation on Burn Wound Healing in Diabetic Rats

摘要


本研究探討波長630 nm 發光二極體(Light-Emitting Diode, LED)光刺激對糖尿病老鼠燒燙傷口癒合之影響。使用Alloxan monohydrate(150 mg/kg, i. p.)藥物進行糖尿病老鼠之誘導。動物於藥物誘導二星期後,利用血糖機測量尾靜脈血液中的葡萄糖含量。葡萄糖濃度≧250 mg/dL 之動物將以Pentobarbital (50 mg/kg, i. p.) 麻醉。再將動物背部上的毛剔除和在背脊左右各1 cm 的位置以溫度100℃的銅片接觸表皮20 sec,每隻動物分別製造四個面積大小各為1×1 cm2 的燒燙傷口。並將動物任意分為二組: 組別I 是未照光組; 組別II 是照光組, 採用波長630 nm 的發光二極體,每二天給予一次光刺激,功率密度為14 mW/cm2,能量密度為4 J/cm2 ,照射時間286 sec。評估方式是採用觀察傷口面積大小、表皮再生、細胞計數和計算嗜中性白血球與巨噬細胞的比率,作為比較光刺激與否對燒燙傷口癒合的影響。實驗 結果發現,光刺激可以加速傷口的癒合,分別在燒燙傷後第5、7、19 和23 天,比較未照光組和照光組的傷口面積大小,並具有統計學上顯著的差異(P<0.01)。組織學評估發現,照光組的傷口表皮再生速率,分別在燒燙後第7、9、15、17 和19 天,都比未照光組的表皮再 生速率要快,並有統計學上的差異(P<0.05)。在細胞計數結果部分,在燒燙傷後第15 和17 天,照光組的巨噬細胞數目比未照光組多。在燒燙傷後第25 天,照光組的血管數目略比未照光組多。但是,比較未照光組與照光組的嗜中性白血球、巨噬細胞、纖維母細胞和血管數 目,在統計學上並無顯著的差異(P>0.05)。在燒燙傷後第25 天,嗜中性白血球和巨噬細胞的比率為0.64,而未照光組的比率為0.78。由以上結果顯示,以波長630 nm 發光二極體,功率密度14 mW/cm2 和能量密度4 J/cm2 照射糖尿病老鼠的燒燙傷口,是可以促進傷口的癒合。

The aim of study was to determine the effect of 630 nm light-emitting diode (LED) photo-stimulation on burn wound healing in diabetic rats. Diabetes was induced in male Wistar rats by single intraperitoneal injection (150 mg/kg of body weight) of Alloxan monohydrate. The blood glucose level was tested two weeks later after Alloxan injection to confirm diabetes (blood glucose greater than 250 mg/dL). After diabetic rats were anesthetized with pentobarbital (50 mg//kg, i. p.), a metal rod pre-heated to 100℃ was used and applied to the dorsal side of the rats for 20 sec. Four burn areas were created and the average size of each burn site was 1×1 cm2. Sham irradiation was applied in the Group I rats as controls. The rats in Group II were irradiated with a dose of 14 mW/cm2 and the irradiation time was 286 sec/treat/every other day. The setting of energy density was 4 J/cm2. The wound healing were evaluated the wound area; re-epithelialization; cell counting and Nutrophils/Macrophages ratio to compare the non-irradiation and irradiation group on wound repair in diabetic rats. In this results showed that irradiation stimulation significantly improved the rate of wound closure in diabetic rats. On post-burn day(PBD) 7、9、15、17 and 19, the histological (Tissue mapping) analysis confirmed the re-epithelialization in rradiation group was faster than that in the controls (P<0.05). The number of macrophages in the wounds treated with irradiation were greater than that in the controls on PBD 15 and 17. And on PBD 25, the blood vessels of counting number in irradiation group were higher than that in non-irradiation group. As compared to the non-irradiation and irradiation groups on the statistics that there were no significant different on the cell counting of neutrophils, macrophages, fibroblasts and blood vessels (P>0.05). The ratio of neutrophils/macrophages of the wound that treated with irradiation was 0.64 and the ratio of non-irradiation group was 0.78 on PBD25. Finally, light-emitting diode irradiation can accelerate the wound healing process in diabetic rats.
