大安水蓑衣(Hygrophilia pogonocalyx Hayata.)組織培養及再生植株之抗氧化能力與抑菌能力分析

摘要


大安水蓑衣(Hygrophila pogonocalyx Hayata)以嫩葉為培植體,於MS培養基添加0.5 mg l-1 BA和0.1 mg l-1 IAA,培養8週後可得22.8±1.9個不定芽,平均芽長 1.0±0.1 公分。不定芽培養於MS培養基,一週後不定芽可再生不定根並發育成小植株。以節段作為培植體,液態靜置培養,每盒接種6-8個培植體,初始鮮重為0.66 g,培養6週後可得9.0±0.1株,平均株高11.6±1.0公分即可出瓶,植株出瓶1個月後,有99.8 %存活率。 再生植株出瓶2個月後,每月採收枝條,以50 %乙醇分別萃取莖與葉,分析粗萃取液抗氧化能力及抑菌能力;11月葉的粗萃取液有最高DPPH自由基清除能力,118.7±3.8 mg ascorbic acid/ g DW ,5月葉的粗萃取液有最高亞鐵離子螯合能力 ( 37.9±0.3 μmole EDTA/g DW );11月葉的粗萃取液有最高總酚含量 (585.5±52.8 mg GA/ 100g DW )。11月份葉的乙醇濃縮液有最好的抑菌作用,對金黃葡萄球菌抑制作用相當於4.79±0.36 μg Ampicillin eq/ g extract,對白色念珠菌抑制作用相當於116.04±4.36 μg Amphotericin B eq / g extract。

The young leaves of Hygrophila pogonocalyx Hayata., were used as explants and cultured on MS medium supplemented with 0.5 mg l-1 BA and 0.1 mg l-1 IAA for organogenesis. The average number of adventitious buds per explant was 22.8±1.9 and the average length of adventitious buds was 1.0±0.1 cm after 8 weeks culture. The adventitious buds rooted and regenerated into plantlet when cultured on MS medium without plant regulator. For mass production of plantlet, six to eight node explants were cultured in sterile vessel (OS14+ODS140,COMBINESS) with 100 ml liquid MS medium. The average number and height of plantlet per vessel were 9.0±0.1 and 11.6±1.0 cm, respectively. The survival ratio of plantlet was 99.8%. Stems were collected monthly after regenerated plantlet acclimatized ex vitro. The antioxidant and antimicrobial activity of ethanol extracts from stems and leaves were investigated. The ethanol extracts showed the highest DPPH radical scavenging activity (118.7±3.8 mg ascorbic acid/ g DW). and expressed the highest ferrous ion chelating activity(37.9±0.3 μmole EDTA/g DW). The ethanol extracts have the highest tatol phenol content (585.5±52.8 mg GA/ 100g DW). The extracts from leaves harvested each month performed antimicrobial activities against Staphylococcus aureus and Candida albicans
