高雄六龜山區臺灣山茶鮮葉之化學成分與抗氧化活性

CHEMICAL COMPOSITION AND ANTIOXIDANT ACTIVITY FROM FRESH LEAVES OF CAMELLIA FORMOSENSIS IN LIOUGUEI MOUNTAIN AREAS

摘要


本試驗針對六龜試驗林不同地區(多納、扇平及南鳳山)臺灣山茶(Camellia formosensis)綠心芽與紅心芽品系鮮葉之熱水抽出物,進行抗氧化活性比較,分析其總酚類化合物含量,並評估其清除DPPH自由基之活性,同時以高效能液相層析儀(HPLC)對各地區及品系鮮葉熱水抽出物之主要多酚類化合物進行定量分析。試驗結果顯示,六龜試驗林不同品系之臺灣山茶皆具有豐富的多酚類化合物及優異的抗氧化活性,其中以扇平之綠心芽與紅心芽品系之總酚類化合物含量最高,分別為445.3 ± 2.5 mg GAE/g與370.9 ± 1.8 mg GAE/g,顯著高於其他地區,並有較佳之清除DPPH自由基能力。根據HPLC分析茶葉多酚類含量,臺灣山茶鮮葉熱水抽出物的成分以兒茶素(-)-EGCG((-)-Epigallocatechin gallate) (96.1 ± 0.1 mg/g - 133.1 ± 2.5 mg/g)及咖啡因(70.7 ± 2.4 mg/g -123.5 ± 3.6 mg/g)含量較高,其中多納地區綠心芽品系之(-)-EGCG顯著高於其他地區,而南鳳山地區則以咖啡因含量顯著高於其他地區。本研究證實六龜試驗林之臺灣山茶具有優異的抗氧化活性,並深具商機潛力。

The objective of this study was to evaluate the total phenolic contents and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity of different cultivars from fresh leaves of Camellia formosensis in Liouguei Experiment Forest (Duona, Shanping, and Nan-Fang Mountain region). In addition, the major phenolic compounds of hot water extracts from various tea specimens were quantified by using high performance liquid chromatography (HPLC). Results revealed that fresh leaves of C. formosensis from Liouguei Experiment Forest were rich in polyphenolic compounds and exhibited great antioxidant activity. Both green (445.3 ± 2.5 mg GAE/g) and red (370.9 ± 1.8 mg GAE/g) sprout cultivars of C. formosensis from Shanping region showed significantly higher content of polyphenolic compounds than the other regions, and had high effectiveness in scavenging DPPH free radical. HPLC analyses revealed that (-)-epigallocatechin gallate ((-)-EGCG) (96.1 ± 0.1 mg/g - 133.1 ± 2.5 mg/g) and caffeine (70.7 ± 2.4 mg/g - 123.5 ± 3.6 mg/g) were the major components in hot water extracts from C. formosensis fresh leaves. The (-)-EGCG contents was significantly higher in green sprout cultivar from Duona region than from the others, while Nan-Fang Mountain region showed significantly higher contents of caffeine than the others. These results demonstrated that the hot water extract of C. formosensis fresh leaves from Liouguei Experiment Forest had an excellent antioxidant activity, and that would have high business potential.
