The Mechanism of LPA-induced IL-1β Expression in J774A.1 Mouse Macrophage Cell Line

摘要


Lysophosphatidic acid (LPA)爲一具有生物功能之水解磷酸酯。其功能包括影響細胞複製、存活,細胞遷移及附著等,又可被視爲一種酯質生長因子。LPA接受器爲G蛋白連結接受器(G protein-coupled receptor),可藉著活化不同的G蛋白來完成LPA對於細胞的影響。IL-1β扮演著促發炎的功能,在非特異性免疫中相當重要。本篇報告的數據顯示,LPA可藉由小鼠J774A.1巨噬細胞株上所表現的LPA接受器,造成細胞內鈣離子濃度上升,並促進IL-1β mRNA及蛋白質的表現。

Lysophophatidic acid (LPA) is a simple lysophospholipid but with complex biological activities. LPA has similar cellular functions with protein growth factors and is viewed as another kind of lipid growth factor. The biological activities elicited by LPA are mediated through the G protein-coupled LPA receptors. IL1-β, a major pro-inflammatory cytokine secreted by many cell types, plays an important role in innate immunity. In this study, we found that LPA induce calcium mobilization and C3-, pertussis toxin-, PDTC- and wortmannin-dependent IL-1β expression in mouse J774A.1 macrophage cell line. These data suggested that LPA is involved in macrophage activation and the process of inflammation.
