Comparison of Luciferase-based Dioxin Detection System Using Fish and Mammalian Cells

摘要


本實驗欲利用花身雞魚的鰭細胞(JF cell),建立改良式的生物細胞戴奧辛偵測系統,達到低成本、高度敏感且可同時篩檢大量樣本的目標。本實驗將已接上luciferase基因和戴奧辛反應因子的受器載體(receptorvector)轉殖入JF cell。經過戴奧辛樣本處理之後,偵測細胞中luciferase冷光酵素的含量,即可反映出受測樣本中戴奧辛的含量。然而,實驗結果顯示JF細胞對於戴奧辛的處理並沒有顯著的反應,似乎無法有效偵測樣品中戴奧辛的濃度;同時,細胞形態在處理前後也沒有顯著的改變。因此我們懷疑JF細胞的質體轉殖效率不甚理想,以致於無法勝任未來生物檢測方法的理想材料。

In this experiment, we attempted to construct an alternative biological detection system based on JF cell as a replacement to mammalian cell system. We expect to be able to construct a low cost, highly-sensitive and efficient dioxin detection tool. The report's vectors were constructed with dioxin response element coupled to luciferase gene. After transfection, we can detect the light intensity in response to dioxin concentration by luminometer and estimate the amount of dioxin presented. However, our results showed that the response of JF cells to dioxin treatment does not effectively reflect the concentration of the toxin. Furthermore, the change in morphology is not significant either. Therefore, JF cells seem to be a poor biodetection system for dioxin.
