小家鼠大腸與盲腸微生物相的遺傳分析

摘要


目前細菌培養技術僅能培養出極少比例的細菌種,對於研究微生物的議題上造成了瓶頸。爲了能有效的研究微生物,微生物學者研發出許多不同種分析方式,而本研究所採用的T-RFLP分析方法不需培養細菌即可分析,是目前研究微生物相最有效率的方法之一。本研究結合了T-RFLP以及DNA定序兩種方法針對16S rDNA試圖分析小家鼠大腸及盲腸道內細菌相的組成,期望建立小家鼠腸胃道細菌相的完整相圖,往後對小家鼠族群分析及其腸胃道細菌共演化的研究將有很大的幫助,而且對人類的腸胃道菌相研究也有幫助。本研究結果顯示大腸盲腸兩者的菌相有明顯的不同。

Current microflora cultivating methods can only retain a very small percentage of microflora species, which has constrained the research of microorganisms. Researchers have developed many different methods to overcome the difficulties. T-RFLP, the method used in this study, is considered one of the most efficient. This study employs both T-RFLP and direct DNA sequencing to analyze the 16s rDNA of microfloras in the large intestine and cecum of a wild house mouse to reveal the microflora composition of the gastrointestines. I found significant differences between the compositions of microfloras in the large intestine and cecum.
