Comparison of Dioxin-Induced Nuclear Translocation AHR between Mammalian and Fish Cells

摘要


本實驗之目的在於測試已建立好的GFP-AHR螢光表現載體在哺乳類細胞(J774A.1)與魚類細胞(JF)能否順利產生GFP-AHR並進行核轉運現象,且加以比較。將GFP-AHR螢光表現載體分別轉殖進入J774A.1與JF細胞後,以戴奧辛處理,透過GFP之螢光觀察AHR的核轉運現象,以推估細胞對戴奧辛的反應。結果發現GFP-AHR螢光表現載體在JF細胞與J774A.1細胞都能發生核轉運現象,又以在JF細胞此現象較爲顯著。

Dioxin-Induced nuclear translocation of GFP-AHR within mammalian (J774A.1) and fish (JF) cell line was tested and compared. J774A.1 and JF cells were transfected with GFP-AHR expression vector. After 4 hours of 10-6M dioxin treatment, the nuclear translocation of GFP-AHR is examined by the fluorescence of GFP and effect of dioxin is evaluated. Dioxin-induced nuclear translocations of GFP-AHR were observed in both types of cells. In comparisons, JF cells seem to response to dioxin better in terms of translocation of AHR.
