咖啡莓果對脂肪肝及相關發炎因子之影響

Effects of Coffeeberry on Fatty Liver and Related Inflammatory Factors

摘要


咖啡莓果(coffeeberry, CB)為製作咖啡的原料,已被證實具有抗氧化、提升膠原蛋白生成、修復老化肌膚及提高人體血液中腦源性神經滋養因子(Brain derived neurotrophic factor, BDNF)等功效,然其延緩肝臟異常病變,如對脂肪肝的效果則尚未明瞭。本試驗之目的在探討咖啡莓果是否可以緩解小鼠脂肪肝的現象,並分析其對發炎反應及肝功能指標之影響。以3月齡雄性老化促進小鼠(senescence accelerated mice P8, SAMP8)為試驗模式,分為添加咖啡莓果不同劑量組,另以不添加咖啡莓果為對照組。12週後將動物犧牲,取其肝臟分析發炎相關之蛋白表現,並以組織切片探討脂肪肝之程度,另取血清進行脂肪及肝指數的檢測。結果顯示,CB可降低發炎相關之訊息路徑nuclearfactor κB(NF-κb)、誘發型一氧化氮合成酶(inducible nitric oxide synthase, iNOS)及環氧化酶-2(cyclooxygenase 2, COX-2)蛋白之表現,提高具有保護肝臟細胞功能之微囊泡的分數,並降低肝功能指標丙胺酸轉胺酶(glutamic pyruvic transaminase, GPT),但對總膽固醇與三酸甘油酯並無顯著影響。綜合以上,推測CB可能透過減少轉錄因子NF-κB,降低iNOS及COX-2所誘導之發炎反應,而達到改善肝功能之效果。

Coffeeberry is the raw material of coffee, and presents beneficial effects on scavenging reactive oxygen species, antioxidantion, enhancing collagen synthesis, repairing aging skin, and increasing blood brain derived neurotrophic factor (BDNF) level. However, the effect of coffeeberry on improving liver disorders, such as fatty liver is still unclear. This study aims to investigate the effect of coffeeberry on lessening the fatty liver, and also evaluate the influences on inflammatory factors. Three month old male senescence accelerated mice-prone 8 (SAMP8) are divided into different doses of coffeeberry and control groups, respectively. Mice are sacrificed after 12 wks feeding, and liver tissue is separated for determining inflammatory proteins. Serum is also collected for evaluating the lipid levels and liver function. The results show that coffeeberry significantly lower the inflammatory signaling factors, including nuclearfactor κB (NF-κB), inducible nitric oxide synthase (iNOS) and cyclooxygenase 2 (COX-2). Furthermore, coffeeberry increased liver micro-vesicles score, reduced GPT level, whereas did not affect the total cholesterol and TG values. Our results demonstrate that coffeeberry is able to improve liver function, possibly through reduces NF-κB, iNOS and COX-2 induced inflammation.
