魚肉硝基呋喃及其代謝物殘留檢測

Detection of Fish Meat Nitrofurans and their Metabolite Residues

富來頓(furazolidone, FZD)、富來他頓(furaltadone, FTD)、硝化富樂遜(nitrofurazone, NFZ)和硝化富蘭音(nitrofurantoin, NFT)等四種硝基呋喃類抗菌劑,因為價格低廉且效果佳,故常被廣泛添加於畜禽及水產養殖飼料中及養殖魚之藥浴,但規定其不得在食用肉品中殘留,惟其在生物體內代謝迅速,且代謝物因和肌肉組織蛋白結合而相當安定,故以檢出代謝物來認定養殖業者是否違規使用。本研究目的是利用液相層析質譜儀(liquid chromatography mass spectrometry, LC-MS)分析養殖魚肉中硝基呋喃類抗菌劑及其代謝物殘留,並以酵素連結免疫吸附分析法(enzyme-linked immunosorbent assay, ELISA)進行定性及定量檢測魚肉中FZD之代謝物AOZ(3-amino-2-oxazolidinone)之殘留量。結果顯示,應用ELISA法可於90 min內完成魚肉中AOZ之殘留檢測,其檢出感度、特異性及再現性均佳,故有可能成為實驗室的初步篩選工具,特別是在大量樣品及在短時間內確定AOZ。

Four nitrofuran antibacterial agents such as furazolidone (FZD), furaltadone (FTD), nitrofurazone (NFZ) and nitrofurantoin (NFT) because of the low cost and good effect, there are often widely added to livestock, poultry, aquaculture feeds and cultured fish drug bathing. However, it is not allowed to remain in the meat, and it is metabolized rapidly in the body and the metabolites are quite stable in combination with proteins of muscle tissue, therefore, by detecting the metabolites can determine whether the aquaculture industry illegally used the nitrofurans. The purpose of this study was to analyze the residues of nitrofuran antibacterial agents and their metabolites in cultured fish meat by liquid chromatography mass spectrometry (LC-MS) and the qualitative analysis and quantitative determination of the residual amount of FZD metabolite AOZ (3-amino-2-oxazolidinone) in fish were executed by enzyme-linked immunosorbent assay (ELISA). The results showed that the ELISA method could complete the residue detection of AOZ in fish meat within 90 min, and the detection sensitivity, specificity and reproducibility were good. So, it was possible to be a preliminary screening tool for the laboratory to determine the AOZ especially when a large number of samples and within a short time.
