摘要


生物正交化學發展至今已有十幾年了,從一開始的施陶丁格反應到較近期的四嗪偶聯反應,該領域的研究使得科學家能透過蛋白質的專一位向修飾,進而了解後轉譯修飾在生物體內所扮演的角色。生物正交化學也在新型藥物(如:抗體-藥物共軛體、醣蛋白疫苗、PEGylated蛋白質藥物)的開發上提供了更多元的製備選擇。本篇文章針對不同生物正交化學做一簡單介紹,並探討該領域對於專一位向性蛋白質修飾的貢獻。

Bioorthogonal chemistry have been developing for more than a decade. From the initial Staudinger reaction to the latest tetrazine ligation, scientists have utilized such chemistry for applications in site-directed protein modifications that in turn led to further insight into the role of post-translational modification in the biological system. Moreover, biorthogonal chemistry has provided more diverse preparation for novel therapeutics such as antibody-drug conjugates, glycoprotein vaccines and PEGylated protein drugs. This review focuses on different types of biorthogonal chemistry and their related applications in site-directed protein modifications.
