毛細管電泳樣品線上濃縮技術及其應用

On-line Sample Preconcentration Technique in Capillary Electrophoresis and Its Application

摘要


自毛細管區帶電泳法的理論發表後,毛細管電泳法至今發展已三十餘年,期間無數的分析方法被開發出來。經過不斷的儀器設計改良,包括與更具靈敏度與應用性的偵測器結合及微小化等技術發展,使毛細管電泳法成為成熟且具有多樣化應用性的分離技術。因毛細管的內徑小,偵測窗光徑短且樣品注入量少,致偵測靈敏度不佳,這也使它的應用受限。線上樣品濃縮技術是解決此問題最重要的技術。本文介紹三種以掃集法為基礎的線上樣品濃縮技術,包括LVSS-ASEI-sweeping、EKS-sweeping及micelle to solvent stacking技術,並探討作用機制及其應用,期盼可以將這些技術應用到更多分析領域中並達到預期成效。

Since the first publication of capillary zone electrophoresis, capillary electrophoresis has evolved for more than 30 years. Innumerable analytical methods have been developed during this period of time. Through continually instrument updated, including the combination of more sensitive and adaptable detection systems and miniaturization, making capillary electrophoresis a fully developed and versatile separation technique. Due to the small inner diameter of the capillary, the short optical length and the small sample size, the concentration sensitivity of capillary electrophoresis technique is poor. This also limits its application in some areas. On-line sample preconcentration technique is the most feasible and simple way to solve this problem. This article describes three sweeping-based on-line sample preconcentration techniques, including LVSSASEI- sweeping, EKS-sweeping, and micelle to solvent stacking technique. Their mechanism of action and applications are also mentioned. It is hoped that these techniques can be applied to more analytical fields and achieve the desired results.
