鹵化反應之綠色化學展望:固態鹵化試劑、鹵化酵素及仿生鹵化催化劑探討

Green Chemistry for Halogenation: Research Development of Solid Halogenation Agents, Haloganases and Biomimetic Halogenated Catalyst Studies

摘要


溴化為有機合成之重要的方法,常用於製備眾多特定官能基之前驅物及作為各種反應之中間體,然而常用的溴化試劑-溴水為高揮發性汙染物,因此本篇統整具有位置選擇性、高產率、易於使用等特性之鹵化試劑發展。此外,本篇探討四種鹵化酵素:HIHPOs(heme iron-dependent haloperoxides)、VHPOs(vanadium-dependent haloperoxidases)、nHIHs(nonheme iron-dependent halogenases)、FDHs(flavin-dependent halogenases),及其活性中心與輔因子對鹵化位置選擇性之效應。最後簡要地統整多種以釩金屬錯合物作為仿生鹵化酵素模型之研究。

Bromination is an important strategy of organic synthesis. It is often used to synthesize various precursors for the preparation of designated functional groups and diverse intermediates. However, volatile bromine as bromination reagent is an environment hazard. In this article, we reviewed the developments of brominating reagents with characteristics of high regioselectivity, high yield, and easy to handle. In addition, we also introduced four halogenases including heme iron-dependent haloperoxidases (HIHPOs), vanadium-dependent haloperoxidases (VHPOs), nonheme iron-dependent halogenases (nHIHs), flavin-dependent halogenases (FDHs), as well as their corresponding active sites and cofactors. Finally, biomimetic studies using vanadium metal complexes as bioinspired halogenases models were briefly summaried.
