桑椹抽出物抑制骨質疏鬆及奶薊萃取物抑制乳癌細胞增生之研究

摘要


骨質疏鬆症及乳癌是高盛行率的疾病,尤其常見於女性,罹病後會對於身心造成傷害,因此,探討如何預防或治療是現今不可忽視的重要議題;然而,目前臨床皆對於此兩種疾病投以藥物治療,雖能有效延緩病症,但副作用卻是難以避免。近年來研究證實,天然物中的功能性成份具有預防或延緩疾病之功能,桑椹已被發現有很好的抗氧化能力,亦可有效抑制多種退化性疾病,而奶薊萃取物具有保肝效果,並透過抑制癌細胞生長及促進其細胞凋亡達到抑制皮膚癌、前列腺癌的功能;目前鮮少有研究探討這類天然物對於骨質疏鬆症及乳癌的影響。 本實驗使用桑椹果實之水萃物 (Mulberry water extract;MWE)探討在骨質疏鬆症的作用;及以高溫水萃的方式所取得的奶薊種子水萃物 (Silybum marianum seed water extract;SMSWE)探討在乳癌中所扮演的角色。結果證實,在去除卵巢大鼠 (Ovariectomic;OVX)在給予不同劑量的MWE後,可以有效增加骨小樑的密度及厚度,並減少骨髓中脂肪空泡數量。結合細胞模式探討後發現MWE可以透過調節成骨細胞及蝕骨細胞的恆定而維持骨骼結構。另外,在動物模式中,給予高劑量的SMSWE可以有效的抑制腫瘤的大小,而在腫瘤切片中亦觀察到SMSWE抑制PLK-1的表現。在細胞模式中,利用不同劑量及處理不同時間的SMSWE發現可以有效的抑制細胞的增生,進一步檢測了調節紡錘體的相關蛋白,結果證實,可以降低PLK-1及EXTAH complex (TPX2、HURP、Eg5、p-Eg5及γ-tubulin)的表現。此外,在癌症基因組圖譜數據庫 (TCGA)的資料比對發現PLK-1與乳癌具高對的相關性。由以上結果證實,善加了解天然物特性並加以利用,可以增加對於疾病的治療或臨床輔助。

Osteoporosis and breast cancer have high prevalence in women. These diseases may cause appearance changes, worsen the quality of life and the mental health of the patients. Therefore, how to prevent or treat them is an important issue. Administration of drug can treat the diseases effectively, but the side effects are difficult to avoid. Recent studies have confirmed that some functional ingredients in natural products could prevent or delay the progression of diseases. Previous researches indicated that mulberry has good antioxidative capacity, which effectively inhibit a variety of degenerative diseases. Many studies revealed that Silybum marianum extract has hepatoprotection effect. Moreover, Silybum marianum can cause skin cancer cell apoptosis and inhibit prostate cancer proliferation. However, there is rare study paid attention to the effects of these natural products on osteoporosis and breast cancer. In this study, we explored the effect of Mulberry water extract (MWE) on osteoporosis and high-temperature water extraction of Silybum marianum seed water extract (SMSWE) on breast cancer. The results confirmed that MWE can effectively increase the density, thickness of trabecular bone and reduce the number of adipocytes in the bone marrow of ovariectomic rats. Taken the in vitro results together, MWE can maintain the skeletal structure via regulating the homeostasis between osteoblasts and osteoclasts. On the other side, SMSWE can effectively reduced tumor size along with inhibiting PLK-1 expression. The inhibitory effect of proliferation may be due to regulating the spindle-related proteins, EXTAH complex (TPX2, HURP, Eg5, P-Eg5 and γ-tubulin). The above results confirmed that MWE can improve the osteoporosis in ovariectomic rats effectively; SMSWE can inhibit breast cancer cells proliferation via inhibiting mitosis. In conclusion, understanding the characteristisms of natural products and applying them well can extend the stretagies in treating diseases or clinical auxillary therapy.
