台灣紅豆杉萃取液具有抗發炎、促進傷口癒合等療效,目前已有高端商業化產品如面膜、肥皂、乳液、修復液等。然而成分並不十分清楚。因此研究的目的在透過各種分析方法來鑑定該樣品所含有的主要成分,並希望以此做為產品品質管控的依據。不過紅豆杉樹過去曾因被提煉出抗癌藥物-紫杉醇而聞名,紫杉醇雖有抗癌效果卻也有較強的毒性副作用,因此鑑定紅豆杉萃取液中是否含有紫杉醇乃首要工作,經由HPLC及MALDI-TOF/MS確認樣品液內不含高毒性的紫杉醇以及經由HPLC及LC/MS確認不含紫杉醇前驅物10-去乙醯巴卡亭III,進而確認其活性成分,先將紅豆杉萃取液進行總酚、總黃酮及總萜的含量分析,其含量分別為452.53±8.85 mg/L、27.00±0.18 mg/L及486.27±26.47 mg/L,接者以HPLC分離紅豆杉萃取液各未知成分,將HPLC的層析過程先分成四段落收集,經由活性測試發現三、四段的成分活性最高,後續藉由半製備HPLC大量收集純化三、四段活性成分並且結合LC/MS、GC/MS、1H-NMR、13C-NMR及FT-IR來對結構進行後續的分析。經過比對後目前已鑑定並定量出活性成分有對羥基苯甲酸7.25±0.22 mg及X compound 10.09±0.24 mg。 關鍵字:紅豆杉萃取液,質譜儀,紫杉醇,對羥基苯甲酸。
Taxus mairei extract has the functions of anti-inflammatory and wound healing, and been applied on facial mask, soap, body lotion and repair lotion in the market. However, the compositions is not clear. The purpose for this research is to identify the ingredients of the sample and set up the standard method for quality control. Taxus mairei is known for taxol, a kind of anti-cancer medicine found in Taxus mairei. Although taxol has the function of anti-cancer, it also has strong toxic side-effect. Since Taxus mairei extract is skin care product, the first job is to make sure no taxol exist inside. On this study, HPLC, MALDI-TOS/MS, and ESI/MS are applied to check these is no toxic taxol and its precursor 10-Deacetyl Baccatin III in Taxus mairei extract. The active constituent of total phenols, total flavonoids, and total terpenes in Taxus mairei extract are measured by tranditional methods and are 452.53±8.85 mg/L, 27.00±0.18 mg/L, and 486.27±26.47 mg/L, respectively. The following is to separate the major peaks inside extrect sample by HPLC/UV. The HPLC chromatogram is devided into four parts to check the most active portion as study priority. It found that the third and forth parts are the most portions. These individual active constituents were collected and analyzed by LC/MS, GC/MS, 1H-NMR, 13C-NMR and FT-IR. It was found that the active constituents are 4-hydroxybenzoic acid and X compound in the amount of 7.25±0.22 mg and 10.09±0.24 mg respectively.
