應用固相微萃取(SPME)技術於土肉桂化學品系鑑定

Chemotype Identification of Indigenous Cinnamon (Cinnamomum Osmophloeum) Using Solid-Phase Microextraction Technique

摘要


為了能準確快速的鑑別土肉桂品系,本研究利用固相微萃取(Solid phase microextraction,SPME)技術從事土肉桂葉部揮發成分之分析及品系之鑑定。以PDMS/DVB吸附纖維進行土肉桂葉部揮發成分吸附,吸附條件之選擇以0.3g之樣本重量、50℃之水浴溫度、5min之水浴加熱時間、15min之吸附時間以及5min之脫附時間所獲得之分析效果最佳。不同地理品系土肉桂葉部揮發成分,以SPME方法直接偵測與水蒸餾法所得精油經氣相層析-質譜儀分析鑑定與群團分析後,結果顯示SPME法與水蒸餾法所獲得之分類結果一致。證實SPME為一快速且能準確分析植物揮發成分的方法,可應用於土肉桂揮發成分之分析與品系鑑定,能迅速辨認品系來源且大幅縮減了分析所需的時間,對土肉桂品系之篩選及應用提供了有利的分析工具。

In this study, SPME fibers coated with PDMS/DVB coupled with gas chromatography/mass spectrometry was used to examine the biogenic volatile organic compounds (BVOCs) from leaves of indigenous cinnamon (Cinnamomum osmophloeum Kaneh.) from various geographical provenances and to identify their chemotypes. The optimum operation conditions obtained were summarized as follows: 0.3g of leaf sample was heated at 50℃ water bath for 5min, which absorbed with SPME for 15 min, and then desorbed for 5min in GC/MS. The BVOCs of indigenous cinnamon leaves analyzed from the SPME method correlated well with those of hydrodistilled essential oils. Furthermore, cluster analysis also showed the same result between SPME and hydrodistillation method for the chemotype identification of indigenous cinnamon. These results showed that the SPME method can be used not only for quickly and accurately analyzing BVOCs of indigenous cinnamon leaves, but also for identifying different chemotypes of indigenous cinnamon.
