本研究旨在探討農桿腫瘤菌(Agrobacterium tumefaciens Smith et Townsend)對檸檬桉(Eucalyptus citriodora Hook.)苗木所引起之癌腫病(crown gall disease)對苗木之生理特性及香茅醛(citronellal)與香茅醇(citronellol)精油含量之效應。未感染農桿腫瘤菌檸檬桉苗木之葉部氣孔密度、氣孔長度及光合作用能力均顯著高於感染者。惟未感染農桿腫瘤菌檸檬桉苗木之香茅醛及香茅醇精油含量則顯著低於感染者。自罹癌腫病檸檬桉苗木之根頸腫瘤組織,成功分離出農桿腫瘤病原菌菌株,並將其接種至無菌培養之檸檬桉試管苗。結果顯示檸檬桉無菌苗經接種農桿腫瘤菌後,在子葉脫落處長出淡綠色腫瘤組織。萃取接種農桿腫瘤菌之無菌苗之精油,並以氣相層析儀分析其成分結果顯示,感染農桿腫瘤菌無菌苗之香茅醛及香茅醇精油含量顯著高於未感染者,顯示農桿腫瘤菌會增加檸檬桉苗木之香茅醛及香茅醇精油含量。
We investigated the effects of Agrobacterium tumefaciens Smith et Townsend on physiological characteristics, and contents of citronellal and citronellol of Eucalyptus citriodora Hook. seedlings. Results showed that the leaf stomatal density, stomatal length, and photosynthesis capacity of non-infected seedlings were significantly higher than the infected ones. However, the citronellal and citronellol contents in stems of infected seedlings were significantly higher than that of the non-infected ones. Sterilized seeds were aseptically culturedand inoculated with A. tumefaciens. The in vitro plantlets developed crown galls after inoculation. Further study revealed that the citronellal and citronellol contents of infected E. citriodora seedlings were significantly higher than that of the non-infected ones. This demonstrated that A. tumefaciens could increase the citronellal and citronellol contents of E. citriodora seedlings.
