摘要


基質輔助雷射脫附游離法(matrix-assisted laser desorption/ionization, MALDI)透過小分子做為基質吸收脈衝雷射光能量,使得分析物脫附並游離成氣態離子,通常結合質譜儀進行樣品分析。儘管MALDI具有軟性游離的特性,但對於鍵結較弱易碎裂的分析物仍無法透過MALDI觀察分子完整的樣貌。因此改良MALDI的游離技術使過程更加溫和成為應用上重要的課題。本文將概述幾個重要的MALDI游離模型,並以其為背景介紹近年來MALDI軟游離技術的發展與應用。

Matrix-assisted laser desorption/ionization (MALDI) employs the small molecules as a matrix to absorb photon energy of the pulsed laser. The analyte is then ionized and desorbed into gas phase, which usually combined with a mass spectrometer for sample analysis. Although MALDI has claimed a soft ionization technique, it is still incapable of observing the intact molecule with weak bonding or labile groups. Consequently, it is essential to improve MALDI technique and make the ionization process milder. The article describes several important ionization models of MALDI mechanism. In addition, we also introduce recent developments of "softer" MALDI technique and the applications.
