以簡易、迅速之薄層層析法確認中藥材牡丹皮(簡稱丹皮)及其製劑中牡丹酚(Paeonol)成分。條件為薄層板:Merck Kieselgel 60 F254,展開溶媒n-hexane/chloroform=1:2,檢測器:紫外燈(254 nm);顯色劑:Vanillin-Sulfuric acid Solution,再以高效液相層析法(條件為層析管:μ-Bondapak C18,移動相,甲醇/水=45:75,流速:1.5ml/min,檢測器:UV254nm)測定Paeonol之含量,用以評估目前台灣多種市售丹皮藥材及含丹皮類製劑之品質,並做為其檢驗方法擬定之參考。甲醇為最佳溶媒,水最差。以甲醇為抽提溶媒測定結果發現市售丹皮中藥材中之Paeonol含量以條狀蘇丹皮之平均含量1.92%為最高,條狀西丹皮之0.23%最低,此法之Paeonol最低檢出量為0.05μg。此法可適用於一般中藥製劑。
Paeonol in Moutan Cortex (Paeonia moutan, Ranunculaceae) and Preparation Containing Moutan Cortex can be identified by thin-layer chromatography (TLC) and quantitatively analysed by high-performance liquid chromatography (HPLC) (μ-Bondapak C18, MeOH/H2O=50:50). With the above HPLC method the content of paeonol in moutan cortices and preparations containing Moutan Cortex commercially available on market were analysed. Their results show that the content of Paeonol of Moutan Cortex seems correlated to its market price, but the paeonol content of preparations are various with prescription of chinese medicine and manufacturer.
