五加皮藥材為五加科落葉小灌木細柱五加Eleutherococcus gracilistylus的根皮,市售藥材常有以香加皮,即蘿摩科植物杠柳Periploca sepium的根皮誤用的情形。本次研究目的即在鑑別藥材及製劑中五加皮及其誤用情形。收集市售五加皮藥材及其製劑並抽取檢體DNA,以標準藥材的DNA序列設計引子,Nested PCR-DNA定序方法進行鑑定。5件五加皮藥材檢體其DNA序列與標準藥材相同,均確認為正品,但序列與Gen Bank上Eleutherococcus koreanus之序列僅有1個鹼基的差異,而與Eleutherococcus gracilistylus有2個鹼基的差異。而5件五加皮中藥製劑中,檢出所含之五加皮成分皆是以香加皮替代使用。
Acanthopanacis Cortex, the dried root cortex of Eleutherococcus gracilistylus, is often adulterated with Periplocae Cortex in market. Periplocae Cortex is the dried root cortex of Periploca sepium and contains toxic chemicals. In this study, we employed the nested PCR followed by DNA sequencing to the identification of Acanthopanacis Cortex in Chinese medicinal preparations. The samples of raw material and preparations were collected from local Chinese pharmacies, and then extracted for total DNA. Two sets of nested PCR primers specific for E. gracilistylus and P. sepium were designed based on the DNA sequences of standard raw materials. By sequences analysis, five samples of raw materials mirrored those of the standards. All five samples were identified as E. gracilistylus. One nucleotide in the sequence of E. koreanus and two in E. gracilistylus from GenBank are different from that of the standard. In five samples of preparations, integrant of Acanthopanacis Cortex was absent and all samples contained Periplocae Cortex.
