二種市售緩釋型氯化鉀錠劑溶離度試驗之比較

Comparison of Two Commercial Potassium Chloride Tablets for Slow-Release in a Vitro Dissolution Test

摘要


市售二種緩釋型氯化鉀錠劑Slow-K和Addi-K在不同條件的溶離試驗中均具有緩慢溶離釋出之作用;且這二種錠劑分別於模擬胃液、模擬腸液及水中均具有緩慢溶離釋出之作用;且這二種錠劑分別於模擬胃液、模擬腸液及水中均具有相似的溶離曲線圖,顯示不同的模擬生理體液對Slow-K和Addi-K的溶離速率影響不大。至於該二種錠劑之溶離釋出動力學模式,經曲線吻合結果顯示,其動力學模式不僅符合一階次動力學模式(First order),亦符合Higuchi模式,但經將溶離釋出速率微分方程處理分析後,發現其溶離釋出動力學模式以一階次動力學模式來描述較適宜。同時該二種市售緩釋型錠劑在任何溶離條件下之溶離釋出參數(K, A2hr和A5hr)經Student's t-test比較後,發現Addi-K之溶離釋出速率較Slow-K為快。

Two commerical slow-release potassium chloride tablets, Slow-K和Addi-K have the characteristics of slow-release in the different dissolution conditions. The two tablets had similar dissolution profiles in simulated intes-tinal fluid (pH 6.8), simulated gastric fluid (pH 1.2) and water; which indicates that, the dissolution rates of the two commercial po-tassium chloride tablets are independent of the pH values of the dissolution medium. The release patterns of potassium chloride fitted the First order model and Higuchi matrix mode. These were distinguished by differential rate treatment, which showed that the release followed the First order pattern. It was found that the dissoluton rate of Addi-K were more rapid than Slow-K in different dissolution conditions.
