柑橘柑果苷之研究(4) 桶柑果皮中柑果苷之抽出與精製

摘要


柑果苷自認爲可治療血管之脆弱性與滲透性(Cpillay fragility and permeability)後已成爲醫藥重要之材料,其衍生物與最後分解物亦可提供化學工業上合成新産品之材料,國外製造柑果苷多自甜橙類之果皮中提取,本省盛産之椪柑及桶柑亦含甚高之柑果苷,而桶柑供罐製年達50萬箱以上;其廢棄之果皮,實有提取利用之價值,本研究採取二處不同採收成熟度之桶柑爲材料,其結果摘述如下: ①烘亁之果皮含柑果苷甚高,但隨菓實採收成熟度而遞減,如10月底採收者約含9.5%,而至3月初僅有4.0%,可知其會計師隨採收成熟度有顯著之變化。 ②商業上柑果苷之抽取,多利用碱液浸出法使果皮之組織先予軟化,讓細胞壁之果膠水解,同時可將所含之柑果苷溶解於碱液中,唯此法不適合於桶柑果皮之提取柑果苷,因其收量甚低,影響提取成本。 ③本研究應用熱甲醇抽取柑果苷效果頗佳,20倍量之甲醇,可得比碱液抽出法高出一倍之收量,且甲醇可收回再供應用,同時可分離出相當量之糖類,對製造成本甚爲有利。抽出柑果苷後之果皮沿可供抽取果膠及最後供充飼料之用。 ④純化柑果苷之方法雖可用如Pyridin, formamide, dimethyl formamied等昂貴溶媒予以純化。但製造成本甚高,且此等溶媒之沸點高,不易回收。本研究採用碱液純化法,可得相當高之純度,其純化品由濾紙層次分析,熔點測定,紫外線吸收光譜及結晶等試驗之結果證明與純品者極爲相似,由此可知利用省産之桶柑果皮抽取柑果苷當極具效果及經濟價值也。

Tonkan (Citrus tankan Hayata) peel contains hesperidin with about four to ten percentage in its dried form. The waste tonkan peel from canneries if used for recovering of hesperidin, the benefit on citrus Industry would be made. An investigation on the extraction and purification of hesperidin was carried. The samples were got from two produced areas in different stages of picking maturity. The result indicates that its physiological function may be important to its hesperidin content as changed with the seasonal variation. The alkaline solution was used for degrading the pectin in the cell walls of tonkan peel in order to dissolve the hesperidin in the aqueous phase, however, this method is not good for isolating the hesperidin from tonkan peel. The benefit on the method of extraction and recovery of the hesperidin by the hot methanol as solvent was based on a higher hesperidin yield with about twice than that by the alkaline solution method. And the used methanol can be recovered and used again; Meanwhile, the about same weight of sugar can be simultaneously separated from hesperidin. On the other hand, the peel residue can be used for extracting pectin and for feed stuff. A good result for purifying hesperidin by alkaline dissolving method was used. It indicates that purified hesperidin with a very high purity as compared with the standard one by the paper chromatoragphy, melting point test, ultraviolet adsorption spectra and crystalligeous test.
