水解對相思樹嫩枝抽出物抗氧化活性及抑制黃嘌呤氧化酶活性之影響

Effects of Acid Hydrolysis on the Antioxidative and Xanthine Oxidase Inhibitory Activities of Acacia Confusa Merr. Twig Extractives

Abstracts


本研究以相思樹嫩枝乙醇抽出物之乙酸乙酯可溶部進行水解,再評估水解產物之抗氧化活性與抑制黃嘌呤氧化酶活性。試驗結果顯示,水解產物之抗氧化能力及抑制黃嘌呤氧化酶活性與水解前抽出物相較下均獲得提升,抑制DPPH自由基及超氧自由基之IC50值分別為5.8及4.2μg/mL,至於抑制黃嘌呤氧化酶活性,其IC50值由原先的26.6μg/mL降為15.5μg/mL。進一步以HPLC分析得知水解產物之主成分為Myricetin及少量Quercetin。試驗結果顯示酸水解確實可切斷醣苷鍵結釋放出配醣基,進而提升抽出物之抗氧化能力及抑制黃嘌呤氧化酶活性。

This study aimed at investigating the effects of acid-hydrolysis on the EtOAc fraction of A. confusa twig extract and evaluating its hydrolyte with antioxidant assays and xanthine oxidase (XO) inhibition assay. After acid hydrolysis, the antioxidant activities were improved with lower IC50 values of 5.8 and 4.2 μg/mL in DPPH and NBT assay, respectively. As for XO inhibition assay, hydrolyte showed better inhibitory action with decreased IC50 value of 26.6 to 15.5 μg/mL. It was demonstrated from HPLC analysis that major myricetin and minor quercetin were obtained in the hydrolyte. These results demonstrated that after acid-hydrolyzing the EtOAc fraction of A. confusa twig extracts the aglycones release from glycosides, thus enhancing the antioxidant activities and XO inhibitory action.
