DNA Sequence-Deduced Amino Acid Sequence of an Unidentified Gap of the Rabbit Muscle Phosphofructokinase

由兔肌磷酸果糖激酶基因序列導引其文獻上懸而未決的胺酸序列缺隙

摘要


利用核酸限制酶剪切選殖法與超音波震碎選殖法,本實驗室已選殖並分析出一段約300bp的磷酸果糖激酶(PFK)基因片段,此片段包括一段可產生29個胺基酸的表現序列、一段含165bp的插入序到及其部份的鄰接表現序列。此基因片段所導引出來的胺基酸序列恰好橫跨一個文獻上懸而未決有關PFK胺酸序列的缺隙(gap)問題,在此缺隙兩邊的胺酸序列與本文之基因序列所導引出來的胺酸序列完全吻合,而且本文所導引出來的胺基酸序列在此缺隙中間有九個胺酸基(IQGL VIIGG)又與文獻上報告的枯草桿菌(Bacillus stearothermophilus) PFK相對應之胺基酸序列具極高的同質性,其在蛋白質的二級結構上亦有相類似的特性。基於以上結果,本文推論由此PFK基因片段所導引出來的胺酸序列確為兔肌磷酸果糖激酶的胺酸序列。

DNA sequence of 300 base pairs harboring the unidentified gap of rabbit muscle phosphofructokinase has been identified among fragments generated by restriction endonu clease digestion and sonication of a purified rabbit phosphofurctokinase gene. This region of DNA contains an exon encoding 29 amino acids, an intron of 165 base pairs and part of a second exon. The DN A deduced amino acid sequence precisely matches the residues of the two regions flanking the gap. In addition, a sequence of nine amino acids, i.e., IQGLVIIGG in the gap is homologous to a corresponding region in Bacillus stearothermophilus phosphofructokinase. The secondary structutal features in this gap are discussed.
