奈米薑黃對脂肪肝及相關調控因子之影響

Effects of Nanolized Curcumin on Fatty Liver and Related Modulators

摘要


薑黃素(Curcumin)具抗氧化和抑制脂質過氧化的效果,然其在體內的吸收率低,使得其生物利用率受到影響。透過奈米技術或許可以通過增加表面積,來提高其生物利用率和增強臨床功效。然而,奈米化薑黃素對減輕肝臟疾病的作用仍不清楚。因此本研究目的在探討奈米薑黃對延緩小鼠脂肪肝進展,及對神經滋養及發炎相關因子之影響。將三月齡的雄性SAMP8小鼠分成對照組和分別補充奈米化薑黃或非奈米化薑黃的兩個實驗組,持續12週。結果顯示,給予小鼠奈米化薑黃與非奈米化薑黃飲食,並不會影響其攝取量,也不會造成生長發育障礙,對肝、腎功能亦無不良影響。與控制組相比,兩組薑黃均有較低的脂肪肝指數,且有降低血清三酸甘油酯之功效。此外,亦能加強肝臟中腦源性神經滋養因子及誘導型一氧化氮合成蛋白質之表現量,其中以奈米化薑黃具更顯著的效果。綜合上述,薑黃可能透過調控神經滋養及發炎相關因子之路徑,而達抑制脂肪肝進展之成效,且薑黃奈米化後能有助於其表現。

Curcumin has beneficial effects of antioxidation and inhibiting lipid peroxidation; however, the bioavailability of curcumin is limited due to its poor in situ absorption. Nanotechnology may improve the bioavailability and enhance the clinical efficacy of materials by increasing their surface area. However, the effect of nanolized curcumin on lessening liver disorders is still unknown. Our aim was to investigate the effects of nanolized curcumin on retarding fatty liver progress, and modulating neurotrophic and inflammatory factors. Three-month-old male SAMP8 mice were divided into a control group, and two experimental groups respectively supplemented with nanolized curcumin or non-nanolized curcumin for 12 weeks. Results showed that food intake, body weight gain, and liver and renal functions did not differ among the groups. Both the nanolized and non-nanolized curcumin groups had lower fatty liver scores and serum triglycerides compared to the control group. Moreover, nanolized curcumin showed better effects of enhancing expressions of brain-derived neurotrophic factor (BDNF) and inducible nitric oxide synthase (iNOS) in the liver. The above evidence demonstrates the beneficial effects of curcumin of improving fatty liver through modulating neurotrophic and inflammatory factors, and nanolized curcumin was shown to be more efficient.
