靈芝酸T抑制A549肺腺癌細胞腫瘤的生長及轉移

摘要


目的:肺癌為發生頻率高且預後差的癌症,其高致死率使它近年來竄升為癌症的首位殺手之一。靈芝在傳統醫藥被認為具有抗癌功效,近年來研究顯示靈芝中的三萜類具有抗癌作用。本研究欲探討由靈芝純化出之三萜類的抗癌效果。 方法:評估靈芝酸T在細胞培養系統中抑制肺腺癌細胞株(A549, PC9, PC9-IR, H1650和H1975)生長的能力,並將A549肺癌細胞接種於免疫缺乏小鼠的皮下,再給予靈芝酸T注射治療,評估靈芝酸T的抗癌效能。 結果:靈芝酸T能夠抑制人類肺腺癌細胞株的生長,而相同濃度的靈芝酸T處理並不影響人類周邊血單核球細胞的存活。動物實驗發現靈芝酸T確實能有效抑制A549肺腺癌腫瘤的生長與癌細胞的肝轉移。 結論:本研究從體外細胞實驗與動物實驗證實了靈芝酸T抗肺腺癌的活性,顯示靈芝酸T具有成為抗癌藥物或癌症治療輔助藥物的潛力。此外也證實靈芝三萜類的抗癌效能。

Objectives: Lung cancer is one of the most common malignant diseases with a dismal prognosis; the high mortality rate has made it one of the leading causes of cancer-related death over the past years. As Lingzhi has long been reputed as anticancer medicinal mushroom in folk medicine and it is known that triterpenoids are the anticancer ingredients of the medicinal mushroom, we are interested in investigating the anti-cancer efficacy of ganoderma triterpenoids. Methods: Anti-proliferation activity of ganoderic acid T (GAT) was studied in human lung adenocarcinoma cell lines A549, PC9, PC9-IR, H1650 and H1975 in cell culture. In vivo anticancer efficacy of GAT was assessed in SCID mice implanted subcutaneously with A549 cancer cells. Results: GAT displayed cytotoxicity on the lung cancer cell lines and exerted no effect on survival of normal human peripheral blood mononuclear cells in cell culture system. In A549 tumor xenograft SCID mouse model, GAT inhibited growth and metastasis of the tumors. Conclusions: GAT is a potential anti-cancer agent or therapeutic drug adjuvant for lung adenocarcinoma. The anticancer activity of triterpenoids from Lingzhi is confirmed.
