There are three different Syva Emit amphetamine assay reagents currently in use in Taiwan, namely: Emit st amphetamine, Emit d.a.u. amphetamine (polyclonal antibody, cutoff 300 ng/ml) and Emit d.a.u. amphetamine/methamphetamine (monoclonal, cutoff 1000 ng/ml). The Emit st reagent is easy to perform, does not require expensive instrument, the personnel does not require extensive training. However, it is inefficient (30 samples per hour) and expensive (260 NT dollars per sample). The Emit d.a.u. polyclonal and monoclonal amphetamine assays can be adapted to automated clinical chemistry analyzer (Hitachi 747). It is very efficient (600 samples per hour, maximum 2400 tests per hour). It wastes about 5 ml of working reagents to prime the instrument. We compared the performance of the Emit st and Emit d.a.u. polyclonal amphetamine reagents and found that these two reagents are comparable. The d.a.u. assays cost only about one tenth of the st reagents. The monoclonal d.a.u. amphetamine assays are more specific, but the separation between the calibrator and the negative sample is smaller than the polyclonal reagent. The d.au. reagents can also be adapted to the microplate reader from the Molecular Devices. Assays performed with plates reader require manual addition of samples and reagents.
