免疫分析之全血液樣本保存

摘要


免疫分析的主要目的在於提供有效數據以解釋說明,因此如何確知全血液樣本在血液組成(Blood composition)、細胞數量(Cell counts)和細胞功能(Cellularfunctions)於保存期間產生了哪些變化,就顯得相當的重要。本篇綜評性論文的主要目的在分析探討如何有效的保存全血液,以獲致精確有效的免疫測量結果,探討內容包括同質性(Homogeneity)、保存期限、溫度、酸鹼度(pH)、血糖濃度、離心(Centrifugation)和振動(Agitation)等變項對全血液保存的影響。理想的全血液樣本的保存須符合下列條件:1.保存在20℃到24℃的溫度環境中。2.血液組成、細胞數量和細胞活性之測量應分別在取樣後48小時、24小時和6小時内完成為宜。3.維持pH介於7.2和7.8之間。4.保持葡萄糖濃度介於100-400 mg/dL(5.5-22.0mmol/L)之間。5.避免振動,但在測量前做充分之和緩多軸的搖晃混合。
