Porphyromonas gingivalis (Pg) is a crucial periodontal pathogen which can destroy the periodontal tissue significantly. The enzyme, gingipain released by Pg, has a great contribution in this effort. Gingipain is a typsin-like enzyme. It can degrade various cytokines or the receptors over immune cells to cause immune suppression. Gingipain can also have influences on blood clotting system; moreover, it will cause connective tissue deconstruction and extensively damage the periodontal tissue. This article is a systemic review of the pathogenic mechanism of gingipain and the role of gingipain in periodontal disease.
